Product Overview:
RIPAb+ antibodies are evaluated using the RNA Binding Protein Immunoprecipitation (RIP) assay. Each RIPAb+ antibody set includes quantitative RT-PCR control primers (RIP Primers) to biologically validate your IP results by successfully coprecipitating a specific RNA target (where such a specific target is known). The qPCR protocol and primer sequences are provided, allowing researchers to validate RIP protocols when using the antibody in their experimental context. Each set also includes a negative control antibody to ensure specificity of the RIP reaction.
The RIPAb+ SNRNP70 set includes the SNRNP70 antibody, a negative control antibody (purified rabbit IgG), and positive qPCR primers which amplify a 100 bp fragment of the human cDNA of U1snRNA. The SNRNP70 and negative control antibodies are supplied in a scalable "per RIP" reaction size and can be used to functionally validate the precipitation of SNRNP70-associated RNA.

Immunoprecipitation from RIP lysate:
RIP lysate from HeLa cells (2 X 106 cell equivalents per IP) was subjected to immunoprecipitation using 0.5 μg of either a normal rabbit IgG or Anti-SNRNP70 antibody. Precipitated proteins were resolved by electrophoresis, transferred to nitrocellulose and probed with anti-SNRNP70 (1.0 μg/mL). Proteins were visualized using One-Step™ IP-Western kit (GenScript Cat. # L00231) (Please see figures).
Control:
Included negative control rabbit IgG antibody and control primers specific for U1snRNA.

RIP Lysate prepared from HeLa cells (2 X 107 cell equivalents per IP) were subjected to immunoprecipitation using 5 µg of either a normal rabbit IgG, or Anti-SNRNP70 antibody and the Magna RIP A Kit (Cat. # 17-700).
Successful immunoprecipitation of SNRNP70-associated RNA was verified by qPCR using RIP Primers U1 snRNA (Please see figures).

One vial containing 50 μg of antibody lyophilized from PBS buffer with 2% sucrose. Add 50 µL of distilled water. Final antibody concentration is 1 mg/mL. For longer periods of storage store at -20°C.

Normal Rabbit IgG
One vial containing 125 μg of purified rabbit IgG in 125 μL storage buffer containing 0.1% sodium azide. Store at -20°C.

RIP Primers, U1snRNA
One vial containing 75 μL of 5 μM of each primer specific for the cDNA of U1 snRNP. Store at -20°C.

Storage Conditions:
Stable for 1 year at -20°C from date of receipt. Handling Recommendations: Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

FUNCTION: Mediates the splicing of pre-mRNA by binding to the loop I region of U1-snRNA. The truncated isoforms cannot bind U1-snRNA.

SUBUNIT STRUCTURE: Found in a pre-mRNA splicing complex with SFRS4, SFRS5, SNRNP70, SNRPA1, SRRM1 and SRRM2. Found in a pre-mRNA exonic splicing enhancer (ESE) complex with SNRNP70, SNRPA1, SRRM1 and TRA2B/SFRS10. Interacts with dephosphorylated FUSIP1 and SFPQ. Interacts with NUDT21/CPSF5, CPSF6, SFRS2IP, and ZRANB2.

SUBCELLULAR LOCATION: Nucleus.

POST-TRANSLATIONAL MODIFICATION: The N-terminus is blocked.
Extensively phosphorylated on serine residues in the C-terminal region.
