Background Information:
Fas (also known as Apo-1 and CD95) is a 40-50 kDa cell surface glycoprotein and a member of the TNF receptor family. Fas is an apotosis signalling molecule that forms the death inducing signalling complex (DISC) upon binding by the Fas ligand trimer on an adjacent cell. The Fas receptor also trimerizes, after which it is internalized and can then bind to the adaptor molecule FADD. FADD facilitates binding of caspase-8. which is activated to induce caspase-8 mediated apoptosis. Fas also plays a role in T cell mediated cytotoxicity and T cell development. Fas is found in the thymus, liver, heart, and ovary, where it is expressed on a minority of resting T and B cells and monocytes. The antagonistic ZB4 clone anti-Fas monoclonal has been used to block the apoptosis-inducing activity of the Anti-Fas CH-11 clone in multiple publications.

Neutralization: 10-500 ng/mL of this antibody was incubated for one hour with human Fas-transfected cells and inhibited the anti-Fas (human, activating), clone CH11 (Catalog # 05-201). Cell viability was assessed using a WST-1 assay

The protein encoded by this gene is a member of the TNF-receptor superfamily. This receptor contains a death domain. It has been shown to play a central role in the physiological regulation of programmed cell death, and has been implicated in the pathogenesis of various malignancies and diseases of the immune system. The interaction of this receptor with its ligand allows the formation of a death-inducing signaling complex that includes Fas-associated death domain protein (FADD), caspase 8, and caspase 10. The autoproteolytic processing of the caspases in the complex triggers a downstream caspase cascade, and leads to apoptosis. This receptor has been also shown to activate NF-kappaB, MAPK3/ERK1, and MAPK8/JNK, and is found to be involved in transducing the proliferating signals in normal diploid fibroblast and T cells. At least eight alternatively spliced transcript variants encoding seven distinct isoforms have been described. The isoforms lacking the transmembrane domain may negatively regulate the apoptosis mediated by the full length isoform.
