Product Overview:

The Guava EasyCyte™ Mini System has all of the functionality of the Guava EasyCyte System but in a single-sample format, making the system an ideal solution for researchers with lower throughput needs.
The Guava EasyCyte Mini System runs all Guava Assays, enabling easy, fast measurements of GFP expression and viability, cell counting and viability, apoptosis, cell cycle analysis, antigen detection, cytotoxicity, and more. Assays require only micro-volumes of cells and reagents, and the system generates minimal waste.
In addition, like all Guava Systems, the unique ability to obtain direct absolute cell counts in addition to population percentages, allows researchers to monitor increases or decreases in overall cell number so it can be determined if they had an impact on the assay being performed. This functionality cannot be obtained with traditional sheath flow cytometers.

Does the system perform color compensation? » Answer

With the fluorescence emission detected in the parameters used for many of the applications on the Guava Systems nearly 100 nm apart, and a selection of fluorochromes with narrow emission peaks, there is less need for compensation. The Guava Express Plus and Guava Express Pro Software Modules (available on most platforms) can perform pre-acquisition compensation for multi-color experiments where it is needed. Also, many third party software packages (such as DeNovo's FCS Express) can perform compensation in analysis mode.

Which fluorochromes can Guava Systems detect? » Answer

Any dyes that excite normally with 532 nm laser light (PCA and PCA-96 System) or 488 nm light (EasyCyte, EasyCyte Plus, or EasyCyte Mini System) and emit in the PMT ranges of the specific platform are acceptable. Many commercially available dyes that work well are listed and described in our fluorochrome dye list document. Please request or download a copy if interested.

How can I print from a Guava System? » Answer

In Analysis mode, each file screen can be printed by clicking the "Print" button option as long as the computer is connected to a printer. In order to preserve specific "screen shots" in either Acquisition or Analysis mode, hold down the "Ctrl” and “Alt" keys and then the "Print Screen" key. This will create a "copy" command. The screen shot can then be "pasted" into Word, Excel, PowerPoint, Paint, or other documents. Screen captures can be further trimmed so individual dot plots can be isolated. In the Guava Express® Pro Software Module for the EasyCyte Plus and EasyCyte Mini Systems, the plots themselves can be copied and pasted into other documents.

Are your files compatible with other cytometry software? » Answer

The files are generated in FCS 3.0 format and can be exported to FCS 2.0 format which is compatible with many third party cytometry post-acquisition analysis software packages.

Can I connect the Guava System to my network? » Answer

Yes, the laptop can be readily connected to your internal network and to the Internet. Since we can help you with training and troubleshooting issues by Internet “desktop sharing” technology, we prefer that your laptop is connected to your network so we can communicate in this fashion.

Can I lock up the computer for security? » Answer

Yes. Each laptop contains an attachment for a security cable lock.

Do I need to use your laptop? » Answer

The laptop is specifically configured and optimized to run the system hardware and software, so Guava Technologies does not recommend use of other computers.

Although capillary-based flow is easier to run than sheath-based flow, can the capillary get clogged? » Answer

The internal dimension of the capillary is 100 microns on a side. Handling large or clumpy cells or samples with a lot of debris predisposes the capillary to clogging. However, all Guava Systems have a Clean and Shut Down Software Module, a Quick Clean Cleaning Cycle, and a Backflush Command that, when regularly used, will keep the capillary open. Use of the Guava® Instrument Cleaning Fluid (ICF) will also keep the capillary free of accumulated cell clumps and subcellular debris often associated with the collection of cell culture samples. If needed, the user can manually syringe water or ICF through the flow cell to clean the capillary. The flow cell is user-replaceable and self-aligning.

How easy is it to use a Guava System? » Answer

Due to its advanced capillary-based flow technology, virtually any scientist or clinician can learn to use a Guava System within one day. Guava Technologies will provide on-site training at the time of installation. You will generally be up and running within a few hours.

How many parameters do Guava Instruments have? » Answer

Depending on the instrument platform, they have from three to six parameters.

What type of laser do instruments have? » Answer

All systems have a solid state diode laser which emits light at 488 nm or 532 nm, depending on the system platform.

Is CytoSoft® Software compatible with Windows™ XP? » Answer

Yes, all current versions of CytoSoft Software are compatible with Windows XP.

Can the software be installed off-line for analysis? » Answer

Yes, Guava CytoSoft™ Software can be installed to run in Analysis mode on computers not connected to the instrument.

Can bacteria be run on the Guava Systems? » Answer

Many strains of bacteria are visible on the Guava EasyCyte Plus System, but Guava Technologies does not, at this time, have a dedicated software interface or protocol for bacteria staining, acquisition, and data interpretation.

Can absolute counts of white blood cells in whole blood be performed with a Guava® System? » Answer

Yes. However, in most cases, an initial dilution step must be performed in buffer such as PBS (Phosphate Buffered Saline) to avoid saturating the system with red blood cell events.

Can the systems perform fluorescence measurements on non-single cell suspensions? » Answer

No, acquisition and analysis of sample requires passage of single cell events through the capillary; however, many enzymatic reagents and protocols exist to disperse solid tissue for proper sample preparation.

Can GFP be detected with Guava® Instruments? » Answer

The fluorophore has minimal signal in the Guava PCA and PCA-96 Systems, but is well-visualized with the Guava EasyCyte Plus and EasyCyte Mini Systems, which have the 488 nm laser.

Can samples be retrieved after acquisition? » Answer

No, the samples pass to the waste vial following acquisition.

Can the system count clumpy cell samples? » Answer

Very clumpy samples can both give inaccurate "undercount" results and clog the flow cell. Cells can be pretreated prior to acquisition with the Guava® Cell Dispersal Reagent (CDR) to alleviate problems caused by chronically clumpy cells.

How many cells do I need for analysis? » Answer

Guava Technologies recommends running samples with a concentration from 10,000 to 500,000 cells/mL. As few as 2,000 cells can be used per sample.

What is the minimum volume required to run an assay? » Answer

The minimum volume of sample required in a microcentrifuge tube for acquisition on the PCA or EasyCyte Mini Systems is 70 microliters. The minimum sample volume required in a microplate well for acquisition on the PCA-96 or EasyCyte Plus Systems is 125 microliters for round-bottom plates and 175 microliters for flat-bottom plates.
