The Guava® Mitochondrial Depolarization Software Module, when used in conjunction with the Guava EasyCyte™ Plus and EasyCyte™ Mini Systems, allows you to quickly differentiate healthy, polarized cells from apoptotic, depolarized cells.
The assay software is unique in that it also allows you to assess if the cells that have entered apoptosis have done so with or without depolarization. This is done by the addition of the dead cell dye, 7-AAD. The software comes with three dot plots that have the X and Y axes labeled to easily identify and gate for the cell populations of interest.
Plot 1: forward scatter vs JC-1 aggregates
Plot 2: JC-1 monomers vs JC-1 aggregates
Plot 3: 7-AAD labeled dead cells vs JC-1 aggregates
The results table automatically displays statistics and direct, absolute cell count information, eliminating the need to perform these statistical analyses yourself. The five different cell populations and their relevant statistics are displayed for easy interpretation of results and include polarized cells, depolarized cells 1, 2, and 3, and apoptotic/dead cells.
