Background Information:
BAX (BCL2-associated X protein) belongs to the BCL2 protein family. BCL2 family members form hetero- or homodimers and act as anti- or pro-apoptotic regulators that are involved in a wide variety of cellular activities. BAX protein forms a heterodimer with BCL2, and functions as an apoptotic activator. This protein is reported to interact with, and increase the opening of, the mitochondrial voltage-dependent anion channel (VDAC), which leads to the loss in membrane potential and the release of cytochrome c. The expression of this gene is regulated by the tumor suppressor P53 and has been shown to be involved in P53-mediated apoptosis. Multiple alternatively spliced transcript variants, which encode different isoforms, have been reported for this gene.

Application Notes:
Immunoprecipitation:
4 μg of previous lots immunoprecipitated Bax from human HL-60 and mouse ABE 8 1/2 lysates.

Immunocytochemistry:
10 μg/mL of previous lots detected Bax in mouse ABE 8 1/2 cells fixed in 1% paraformaldehyde, followed by permeabilization with 100% methanol for 60 seconds.33

Immunohistochemistry (Paraffin):
Paraffin embedded, well-differentiated ductal adenocarcinoma, stained with anti-Bax NT (10 μg/mL)
Control:
Human heart tissue, human kidney tiussue, human placenta tissue, HepG2 cell lysate.
Quality Assurance:
Routinely evaluated by western blot on cell lysates from human HL-60 cells.

Western Blot Analysis: 0.5-2 μg/mL of this lot detected Bax in 20 μg of cell lysates from human HL-60 cells.

Purified rabbit IgG in buffer containing 0.1 M Tris-glycine, pH 7.4, 0.15 M NaCl, 0.05% sodium azide before the addition of glycerol to 30%.
Storage Conditions:

Handling Recommendations:
Upon receipt, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C.
Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
Note: Variability in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.
