Anti-iNOS/NOS II, NT

Species Reactivity Key Applications Host Format Antibody Type
M, B, Sh, Gp IH, IP, WB Rabbit Purified Polyclonal Antibody
Description:
Anti-iNOS/NOS II, NT
Trade Name:
Upstate (Millipore)
Product Overview:
Note: ADMA (Catalog #19-169) and NMMA (Catalog #19-170) are inhibitors of NOS
Specificity:
Recognizes N-terminus of iNOS, Mr 125 kDa. Does not cross-react with nNOS/NOS I.
Molecular Weight:
125 kDa
Epitope:
N-terminus
Immunogen:
Partial fusion protein raised against the N-terminus of murine iNOS/NOS II.
Isotype:
Background Information:
Nitric oxide (NO) is an inorganic, gaseous free radical that carries a variety of messages between cells. Vasorelaxation, neurotransmission and cytotoxicity can all be potentiated through cellular response to NO. NO production is mediated by members of the nitric oxide synthase (NOS) family. NOS catalyzes the oxidization of L-arginine to produce L-citrulline and NO. Two constitutive isoforms, brain or neuronal NOS (b or nNOS, type I) & endothelial cell NOS (eNOS, type III), and one inducible isoform (iNOS, type II), have been cloned. Cytokines such as interferon-gamma (IFN), tumor necrosis factor (TNF), interleukin-1 and -2, and lipopolysaccarides (LPS) cause an increase in iNOS mRNA, protein, and activity levels. Protein kinase C-stimulating agents exhibit the same effect on iNOS activity. Human iNOS is regulated by calcium/calmodulin (in contrast with mouse NOS2).
Species Reactivity:
  • Mouse
  • Bovine
  • Sheep
  • Guinea Pig
Species Reactivity Note:
Mouse, bovine, sheep and guinea pig. Other species cross-reactivity is unknown.
Application Notes:
Immunohistochemistry:
A previous lot of this antibody has been reported to detect iNOS/NOS II in 2% paraformaldehyde-fixed sheep lung (Sherman, T.S., 1999).

Immunoprecipitation:
4 μg of a previous lot immunoprecipitated iNOS/NOS II from 500 μg of L929B cells which had been stimulated with 10 ng/mL IFNg and 1 μg/mL LPS.
Control:
INFγ and LPS treated L929B cell lysate, cytokine stimulated A549 cells, cytokine stimulated rat fibroblasts
Quality Assurance:
Routinely evaluated by immunoblot on lysates from L929B cells that had been stimulated IFNγ and LPS.

Western Blot Analysis: 1-4 μg/mL of this lot detected iNOS/NOS II in RIPA lysates from RAW 264.7 cells that had been stimulated with LPS. 2 μg/mL of a previous lot detected iNOS/NOS II from similarly treated L8 cells.
Purification Method:
Protein A purfied
Presentation:
Purified IgG in buffer containing 0.02 M phosphate buffer, pH 7.6, 0.25 M NaCl, and 0.1% sodium azide. Frozen solution.
Storage Conditions:
Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variability in freezer temperatures below -20°C may cause glycerol-containing solutions to become frozen during storage.

Entrez Gene Summary:
Nitric oxide is a reactive free radical which acts as a biologic mediator in several processes, including neurotransmission and antimicrobial and antitumoral activities. This gene encodes a nitric oxide synthase which is expressed in liver and is inducible by a combination of lipopolysaccharide and certain cytokines. Three related pseudogenes are located within the Smith-Magenis syndrome region on chromosome 17.
UniProt Summary:
FUNCTION: SwissProt: P35228 # Produces nitric oxide (NO) which is a messenger molecule with diverse functions throughout the body. In macrophages, NO mediates tumoricidal and bactericidal actions.
COFACTOR: Heme group (By similarity). & Binds 1 FAD (By similarity). & Binds 1 FMN (By similarity). & Metrahydrobiopterin (BH4). May stabilize the dimeric form of the enzyme (By similarity).
