Anti-RAD50

Species Reactivity Key Applications Host Format Antibody Type
M, H, R IC, WB Rabbit Affinity Purified Polyclonal Antibody
Description:
Anti-RAD50
Specificity:
This antibody recognizes RAD50.
Molecular Weight:
~154 kDa
Immunogen:
Synthetic peptide corresponding to RAD50.
Background Information:
RAD50 (previously known as DNA repair protein RAD50) is homologous to Saccharomyces cerevisiae Rad50 and is a member of the structural maintenance of chromosome (SMC) family and is involved in cell growth and viability. RAD50 is known to encode 2 distinct proteins. The RAD50, XRS2/NBS1 complex is required for activation of ATM which will phosphorylate TERF1, thereby promoting telomerase. Along with MRE11 and NBN, RAD50 also forms the MRE11–RAD50–NBS1 (MRN) complex, which is one of the first factors to be localized to the DNA lesion, where it might initially have a structural role by tethering together, and therefore stabilizing, broken chromosomes. MRN also controls ATR, which is required for SMC1A phosphorylation, resulting in chromatid adhesion and DNA damage response. RAD50 is expressed in all tissue but predominantly in the testis and fibroblasts.
Species Reactivity:
  • Mouse
  • Human
  • Rat
Species Reactivity Note:
Tested on human. Predicted to react with mouse and rat based on sequence homology
Application Notes:
Immunocytochemistry:
A 1:500 dilution of this antibody was used to detect RAD50 on HeLa and A431 cells.
Control:
Human spleen lysate.
Quality Assurance:
Western Blot Analysis:
A 1:500 dilution of this antibody was used to detect RAD50 on human spleen lysate.
Purification Method:
Affinity Purfied
Presentation:
Purified rabbit polyclonal antibody precipitated in a solution of 50% saturated ammonium sulfate and PBS containing no preservatives.
Storage Conditions:
Stable for 1 year at 2-8ºC from date of receipt.
To reconstitute the antibody, centrifuge the antibody at a moderate speed (5,000 rpm) for 5 minutes. Carefully remove the ammonium sulfate/PBS buffer solution and discard; 10μL of residual ammonium sulfate solution will not affect the re-suspension of the antibody. Do not let the protein pellet dry, as severe loss of antibody reactivity can occur. Re-suspend the antibody pellet in 100L either standard PBS or TBS (pH 7.3-7.5). DO NOT VORTEX. Mix by gentle stirring with a wide pipet tip or gentle finger-tapping. Let the precipitated antibody rehydrate for 1 hour at 4-25°C prior to use. Small particles of precipitated antibody that fail to re-suspend are normal. Vials are overfilled to compensate for any losses.

UniProt Summary:
FUNCTION:Component of the MRN complex, which plays a central role in double-strand break (DSB) repair, DNA recombination, maintenance of telomere integrity and meiosis. The complex possesses single-strand endonuclease activity and double-strand-specific 3'-5' exonuclease activity, which are provided by MRE11A. RAD50 may be required to bind DNA ends and hold them in close proximity. This could facilitate searches for short or long regions of sequence homology in the recombining DNA templates, and may also stimulate the activity of DNA ligases and/or restrict the nuclease activity of MRE11A to prevent nucleolytic degradation past a given point. The complex may also be required for DNA damage signaling via activation of the ATM kinase. In telomeres the MRN complex may modulate t-loop formation. Ref.5 Ref.6 Ref.7 Ref.11 Ref.13 Ref.17

Cofactor Binds 1 zinc ion per homodimer By similarity.

SUBUNIT STRUCTURE: Component of the MRN complex composed of two heterodimers RAD50/MRE11A associated with a single NBN. Component of the BASC complex, at least composed of BRCA1, MSH2, MSH6, MLH1, ATM, BLM, RAD50, MRE11A and NBN. Found in a complex with TERF2. Interacts with RINT1. Interacts with BRCA1 via its N-terminal domain. Interacts with DCLRE1C/Artemis. Ref.1 Ref.8 Ref.12 Ref.16 Ref.19

SUBCELLULAR LOCATION: Nucleus. Telomere. Note= Localizes to discrete nuclear foci after treatment with genotoxic agents. Ref.8 Ref.9

TISSUE SPECIFICTY: Expressed at very low level in most tissues, except in testis where it is expressed at higher level. Expressed in fibroblasts. Ref.1

DOMAIN:The zinc-hook, which separates the large intramolecular coiled coil regions, contains 2 Cys residues that coordinate one molecule of zinc with the help of the 2 Cys residues of the zinc-hook of another RAD50 molecule, thereby forming a V-shaped homodimer. The two heads of the homodimer, which constitute the ATP-binding domain, interact with the MRE11A homodimer By similarity.

PTM: Phosphorylated upon DNA damage, probably by ATM or ATR By similarity.

MISCELLANEOUS: In case of infection by adenovirus E4, the MRN complex is inactivated and degraded by viral oncoproteins, thereby preventing concatenation of viral genomes in infected cells.

SEQUENCE SIMILARITY:Belongs to the SMC family. RAD50 subfamily.

Contains 1 zinc-hook domain.

SEQUENCE CAUTION: The sequence AAH62603.1 differs from that shown. Reason: Miscellaneous discrepancy. Contaminating sequence. Potential poly-A sequence.
