Background Information:
Methylation of Lysine 79 in Histone 3 acts as a marker of inactive chromatin regions. Methylation by Dot1 methylase on lysine-79 (Lys79) of Histone H3 is criticial for transcriptional silencing, and it is thought that silencing proteins such as Sir3 function by blocking Dot1 methylation. Likewise, it is predicted that acetylation of Lys79 may indicate active gene transcription.

Application Notes:
Western Blot Analysis: A 1:1000-1:2000 dilution detected acetyl-Histone H3 (Lys79) in acid extracts from HeLa cells. Signal is increased in cells treated with sodium butyrate (Catalog # 19-137), a histone deacetylase inhibitor. No detection of recombinant, unmodified human Histone H3 (Catalog # 14-494) was observed.
Optimal working dilutions must be determined by the end user.

Purified rabbit IgG in 70% storage buffer containing 0.1 M Tris-glycine, pH 7.4, 0.15 M NaCl, 0.05% sodium azide, 5 mg/mL BSA and 30% glycerol.
Storage Conditions:
Stable for 1 year at -20°C from date of receipt. Avoid repeated freeze/thaw cycles.
Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variability in freezer temperatures below -20°C may cause glycerol-containing solutions to become frozen during storage.

Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Two molecules of each of the four core histones (H2A, H2B, H3, and H4) form an octamer, around which approximately 146 bp of DNA is wrapped in repeating units, called nucleosomes. The linker histone, H1, interacts with linker DNA between nucleosomes and functions in the compaction of chromatin into higher order structures. This gene contains introns and its mRNA is polyadenylated, unlike most histone genes. The protein encoded is a replication-independent member of the histone H3 family.
