Background Information:
In this homogeneous assay, a phosphorylated fluorescent substrate is used to assay Phosphatase activity. In the phosphorylated state, the fluorescence emission from the substrate is quenched. Once de-phosphorylated by an active Phosphatase, the quench is released and fluorescent emission increases in accordance with the number of de-phosphorylation events. The assay is readily amenable to high-throughput screens, requiring only a phosphatase specific base buffer, in which the substrate and phosphatase are diluted. The assay can be started by adding the substrate or enzyme. The progress of the reaction can be followed either in real-time for kinetic measurements, or in an end-point format. A de-phosphorylated substrate is provided for use in generating a standard curve, enabling quantitative analysis and comparisons of enzymatic activity by treatment condition.
