Product Overview:
The GenoStat Ligand facilitates the control of exogenous gene production when utilizing the GenoStat Inducible Expression System (cat. no. 17-428). This is accomplished through the highly specific binding interaction between the GenoStat Ligand and the bipartite nuclear receptor complex, which is expressed from the pGS-1 Receptor plasmid (cat. no. 21-208). The binding of ligand causes a conformational change in the receptor complex which rapidly activates transcription of an exogenous gene located on the pGS-2 Expressor plasmid (cat. no. 21-209). Thus, the GenoStat Ligand can rapidly induce the expression of the particular gene of interest. Furthermore, this expression can be up regulated by the addition of higher ligand concentrations and repeatedly turned 'on' and 'off' by the addition and removal of ligand.

Technical Information:
The GenoStat™ Ligand is classified as a synthetic diacylhydrazine [(N-(2-ethyl-3-methoxybenzolyl)-N'-(3,5-dimethylbenzoytert-butylhydrazine] and it has a molecular weight of 382.5. As such, it is a member of a family of compounds that function as gene inducers and act as non-steroidal ecdysone agonists.

Application Notes:
The GenoStat™ Ligand is provided at a concentration of 5 mM in tissue culture grade 100% DMSO and any subsequent dilutions of it should also be made using tissue culture grade 100% DMSO. For long term storage the ligand and any dilutions of it should be kept in a -20°C freezer. For short term use, the GenoStat™ Ligand can be added to cell culture media (at the desired final concentration) and stored at 4°C for several weeks with no effect. Furthermore, media containing the ligand can be added to cells in culture and incubated at 37°C for many days while still retaining full functionality. Please note that high concentrations of DMSO in media can adversely affect cell health therefore it should not exceed a final cell culture concentration of 0.5%. The GenoStat™ Ligand can be effectively used at doses in the picomolar to micromolar range, although at final concentrations higher than 500 nM the induction response may not be linear. The optimal dose for maximal gene expression is cell type specific and should be determined by assaying a range of ligand dose responses. Inducible gene expression using this system has been shown to function in numerous different cell lines from various species including human, rat, mouse, and hamster.
