Product Overview:
The GenoStat&rade; 293 Stable Cell Line was developed to help facilitate the expression of exogenous genes when utilizing the GenoStat Inducible Expression System (cat. no. 17-428). This cell line has stably integrated the pGS-1 Receptor plasmid (cat. no. 21-208) into its genome. This was accomplished by transfecting human epithelial kidney 293 cells with the pGS-1 Receptor plasmid (which contains a neomycin selection cassette) and then selecting for stable clones following the addition of neomycin (G418) into the media. Individual clones were isolated, grown under selective pressure, and subsequently assayed for induced gene expression by measuring firefly luciferase activity following the transient transfection of each clone with the pGS-Luciferase reporter plasmid (cat. no. 21-210). The clone showing the highest expression level with the lowest background was selected.

Application Notes:
These 293 stable cells will facilitate the inducible expression of exogenous genes when used in conjunction with the pGS-2 Expressor plasmid (cat. no. 21-209). The end user can simply subclone their gene of interest into the pGS-2 Expressor plasmid and then transfect it into these stable 293 cells. Following transfection, GenoStat™ Ligand (cat. no. 20-333 or 20-382) is added to the culture media to turn "on" expression of the subcloned gene. The expression can be up regulated by the addition of higher concentrations of ligand. Genes can be repeatedly turned "on" and "off" by cycling the addition and removal of ligand. Hence, the utility of this cell line is the reproducibility of the pGS-1 Receptor protein components and the avoidance of co-transfection experiments.

Sterile technique is absolutely required for all cell culture procedures described in this manual. The cryopreserved cells provided should be used to establish cultures immediately upon receipt. Culturing of these cells should be done in a standard 37°C incubator boosted with 5% CO2. As soon as possible after initiating cell cultures, numerous vials of cells should be back-frozen. Sensitivity and responsiveness to ligand may decrease over time, hence it is important to use and store the earliest possible passages of cells. Storage of all cryopreserved cells should be in the vapor phase of liquid nitrogen. Finally, we recommend the use of the associated reagents and procedures described herein to optimize cell line viability and functionality.
