Product Overview:
CHEMICON's Mouse-to-Mouse IHC Detection kit is designed for staining mouse or rabbit antibodies on mouse or rat tissues. The kit utilized a drastically different new technology to eliminate background problems. The system is biotin-free and more sensitive then other similar systems that are available.

For use with mouse primary antibodies on mouse tissues to prevent the background often associated with mouse-on-mouse immunohistochemistry.

Application Notes:
Immunohistochemistry: (All procedures should be performed at room temperature)
Deparaffinization, rehydration
Block endogenous peroxidase with 3% H2O2 in dd water for 10 minutes.
Perform antigen retrieval or enzyme digestion if necessary.
1.Block with Pre-antibody Blocking solution for 60 minutes. Wash with TBS for 5 minutes, two times.
2.Incubate the mouse or rabbit primary antibody for 30-60 minutes, wash with TBS for 5 minutes, two times. (Incubation times with the primary antibody will need to be determined by the end user.)
3.Block with Post-antibody Blocking solution for 10 minutes. Wash with TBS for 5 minutes, two times
4.Incubate with the ready-to-use Poly-HRP-Anti-Mouse-Rabbit IgG for 10 minutes, wash with TBS for 5 minutes, two times.
5.Apply AEC for 10 minutes. Wash with distilled water.
6.Counterstain with hematoxylin and mount.
NOTE:
1.Blocking for >60 minutes may be required for certain tissues (tonsil, lymph node, etc).
2.DAB can be used to replace AEC as substrate if background is clean and higher sensitivity is needed.
3.AEC staining is organic soluble and should be mounted in an aqueous mounting media.
4.The system is biotin free so that blocking endogenous biotin is not necessary.
