How does the Beadlyte system work? » 回答

The Beadlyte system utilizes bead set that are internally labeled with two fluorescent dyes (one for identification of the bead and one for fluorescent intensity determination). This bead set contains surface Carboxyl groups for covalent attachment of ligands (or biomolecules). Once an antibody is conjugated to the bead, a sample is added (ie; lysate), and that is followed by a reporter antibody; which is often biotinylated. Following incubation with the biotinylated reporter antibody, strepavidin/phycoerythrin is added to make detection possible. Phycoerythrin excitation is acheived with a 532nm laser with emission at 550-600nm. The actual Luminex machine utilizes a 563-587nm band pass filter. Unfortunately, the composition of the dyes that label the beads is proprietary.

In regards to the software, are there any advantages of reading a plate with a high RP1 value? » 回答

Reading a plate using “high” RP1 targets may provide increased detection in some assays. If using the higher calibration settings, we recommend that the assay first be read based on the low target value, then re-read based on the high target value. This will prevent the loss of data that may be in the high range of the assay.

In regards to the software, are there any advantages to a high calibration? » 回答

Experiments using Beadlyte cytokine detection systems have shown that the high calibration does not increase sensitivity. Although the MFI values are higher, the background also increases.

In regards to the software, what is the CAL2 setting? » 回答

The CAL2 setting sets the voltage of the photomultiplier tube (PMT) for the laser that detects the reporter.

In regards to the software, what is the CAL1 setting? » 回答

The CAL1 setting sets the voltage of the photomultiplier tube (PMT) for the laser that detects bead colors and defines the bead number.

In regards to the software, what is the doublet discriminator gate? » 回答

The doublet discriminator gate is automatically set at 8,000-13,500 and will exclude doublets or other larger bead aggregates.

Can I use Upstate's Beadlyte products with a BioPlex machine? » 回答

Upstate's Beadlyte product line is fully compatible with the BioPlex machine. Despite differences in the software, the hardware is identical.

What types of samples have been tested in the Beadlyte kits? » 回答

Plasma, serum, tissue culture media, lavage fluid, urine and breast milk have all been used in the Cytokine/Chemokine kits. Cell lysates have been used in the Cell Signaling Kits.

Do I have to dilute my samples in assay buffer? » 回答

It is important to dilute samples and standards in assay buffer which contains BSA. A lack of BSA or low amounts of BSA can affect the curve.

What is the difference between the Luminex 100 and the Luminex 200 machine? » 回答

For the most part, one of the differences between the Luminex 100 and the Luminex 200 machines is that a thumbwheel has been installed for probe height adjustment in the 200 versus a hex screw in the 100.

Can the plates used in the Beadlyte kits be used with a standard ELISA plate washer? » 回答

No. The washes need to occur via a vacuum filter.

What size are the beads? » 回答

The beads are 5.6 microns.

What is the pore size of the 96 well plate? » 回答

The pore size of the 96 well plate is 1.2 microns.

If I am using a plate with a 96 well format, do I have to use the whole plate? » 回答

No. If it is a strip well plate, remove the wells you do not intend to use. If it is not a strip well plate, tape off any unused wells with lab tape to avoid contamination.
