The Guava® Cell Growth Assay provides, at the single-cell level, proliferation assessments out to five generations with a simple, non-radioactive assay that is superior to population-based methods such as 3-Thymidine and MTT (or XTT). In addition, the assay offers a broader linear range when compared to colorimetric methods.
Simple, non-radioactive protocol
The assay uses a well-characterized fluorescent dye, carboxyfluorescein diacetate succinimidyl ester (commonly referred to as CFSE), that diffuses freely into cells and is retained within the cell without affecting cellular function. The dye is not transferred to adjacent cells. For each round of cell division, the relative fluorescence intensity of the dye is decreased by half. A dead cell dye, PI, is included to discern the percentage of cells that are dead.
The assay is ideal for primary cultures such as peripheral blood mononuclear cells (PBMCs), activated T- or B-cells, peripheral blood lymphocytes (PBLs), thymocytes, hematopoietic cells, and splenocytes. For use with EasyCyte, EasyCyte Mini, EasyCyte Plus.
