Product Overview:
The Beadlyte® Phospho-ZAP-70 Beadmate pair is the optimal antibody pair for detection of phosphorylated ZAP-70. Beadmate antibody pairs are used in conjunction with the Beadlyte® Cell Signaling Buffer Kit (Catalog # 48-600, or 48-601) to detect the presence of phosphorylated ZAP-70 in cell lysates using the Luminex® xMAP system. Each Beadmate pair is ordered individually and can be combined for simultaneous multiplex analysis of cellular events. The Beadlyte® Cell Signaling Buffer Kit is also ordered separately and consists of a common set of reagents needed for using Beadmates. The detection assay is a rapid, convenient alternative to Western Blotting and immunoprecipitation procedures. Each kit contains sufficient reagents for 100 individual assays.

Descriptive Text:
ZAP-70 is a non-receptor tyrosine kinase of the Syk family. It is a key early signaling mediator in T-cell activation and is normally only expressed in T-cells; expression in B-cells is a marker for poor prognosis in chronic lymphocytic leukemia. ZAP-70 becomes activated following ligation of the T-cell Receptor. It has two n-terminal SH2 domains, which bind ITAM motifs. When ZAP-70 is bound to an ITAM, Lck trans-phosphorylates and remains associated with it. Downstream effectors of the ZAP-70/Lck complex include LAT, Phospholipase Cγ, Vav, and the Rho and Ras GTPases. Autophosphorylation of ZAP-70 on Tyr319 is critical for association with Lck and thus for activation of downstream effectors.
