Product Overview:
The MILLIPLEX MAP Phospho NFĸB (Ser536) MAPmates pair is used in conjunction with the MILLIPLEXMAP Cell Signaling Buffer and Detection Kit (Catalog # 48-602) to detect the presence of phosphorylated NFĸB (Ser536) in cell lysates using the Luminex® 100™ IS, 200™, or HTS system. Each MAPmates pair is ordered individually and may be combined for simultaneous multiplex analysis of cellular events. The MILLIPLEX MAP Cell Signaling Buffer and Detection Kit is ordered separately and consists of a common set of reagents needed for performing MAPmates assays. The detection assay is a rapid, convenient alternative to Western Blotting and immunoprecipitation procedures. Each kit contains sufficient reagents for 100 individual assays. The MILLIPLEX MAP HeLa Cell Lysate: Unstimulated included with this MAPmates pair may be utilized as a negative control for this target, while the HeLa Cell Lysate: TNFα + calyculin A included with this MAPmates pair may be utilized as a positive control.

Nuclear factor ĸB (NFĸB)
• Transcription factor
• In its Inactive form NFĸB is bound to IĸB in the cytosol
• Stimuli (cytokines, LPS, DNA damage, viral infections, etc.) lead to phosphorylation of IĸB, causing dissociation of NFĸB, which then enters the nucleus and acts as a transcription factor
• Involved in many biological processes, such as inflammation, immunity, differentiation, cell growth, tumorigenesis and apoptosis.

• This assay requires 25 μL diluted cell lysate (recommended starting concentration is 20 μg cell lysate/well)
• This is an overnight (4°C) or two-hour (RT) incubation assay; however, for best results an overnight incubation is recommended.
• MAPmate is compatible with both Assay Buffer 1(Catalog # 43-010) and Assay Buffer 2 (Catalog # 43-041)
