Product Overview:
Cells respond to their environment in many different ways through intracellular signaling. Signaling through Receptor Tyrosine Kinases (RTKs) often promote increased metabolism and cell growth. Epidermal Growth Factor Receptor (EGFR) is a 175 kDa receptor activated by such ligands as EGF and heregulin. EGF binding induces EGFR autophosphorylation at specific tyrosine residues, promoting the docking of several SH2 domain-containing adaptor proteins to EGFR that, in turn, activate downstream signaling pathways such as MAPK, AKT and STAT. In addition to autophosphorylated tyrosine residues, EGFR contains other phosphorylation sites including phosphothreonine and phosphoserine residues that are involved in crosstalk with other signaling pathways and EGFR down regulation.

• An overnight (4°C) incubation is recommended for best results.
• This assay requires 25 µL diluted cell lysate per well.
• This kit must be run using Assay Buffer 1 (provided).
• 1 - 25 µg cell lysate/well (recommended starting concentration is 40 to 1,000 µg protein/mL).
