Background Information:
Nerve Growth Factor Receptor, also termed NGFR p75, p75NTR, or CD271 is the low affinity NGFR (LNGFR) which binds NGF and other neurotrophins including BDNF, NT3, NT4/5 with similar, low affinity. NGFR p75 is a 75 kDa transmembrane glycoprotein (399 a.a.) consisting of an extracellular domain (222 a.a.) which contains four cysteine rich domains responsible for ligand binding, a transmembrane domain (22 a.a.), and a cytoplasmic domain (155 a.a.). NGFR p75 is mainly expressed in Schwann cells and neurons and in a variety of non neuronal cells. NGFR p75 is necessary for regulating neuronal growth, migration, differentiation and cell death during development of the central and peripheral nervous system. NGFR p75 plays a central role in the regulation of cell number by apoptosis in the developing CNS.

Species Reactivity Note:
Rat and chicken. Predicted to react with 100% mouse and human based on sequence homology. It has been reported that this antibody does not recognize human or rat p75 (See MAB365) by Western blot.
Application Notes:
Western Blot:
1:1,000-1:2,000 dilution of a previous lot was used on western blot.

Immunohistochemistry:
1:50 and 1:300 dilution from a previous lot detected NGF Receptor P75 using IHC-Select Detection with HRP-DAB.

Immunohistochemistry(paraffin):
1:150 dilution of a previous lot was used (Bouin’s fixative). Reactive on paraffin embedded sections.

Flow Cytometry:
1:1,000 dilution of a previous lot was used on flow cytometry.

Blocking Assay (Inhibition):
A previous lot of this antibody bound NGF to cells expressing p75 at 1:1,000 dilution.

Immunocytochemistry:
1:500 dilution of this antibody from a previous lot detected NGF Receptor P75.

Immunoprecipitation:
A previous lot of this antibody was used on Immunoprecipitation. 1:500 - 1:5,000 dilution of this lot detected NGF Receptor P75 on 10 µg of PC12 cell lysate.
