Species Reactivity:
- Human
- Rat
Species Reactivity Note:
The epitope specific for Aquaporin 1 is identical in rodent, bovine, sheep and only slightly different in frog (Rana esculenta) (18/19).
Application Notes:
Western blotting: 1:400-1:800 using ECL on rat kidney membranes.
Immunohistochemistry
Dilutions should be made using a carrier protein such as BSA (1-3%).
Optimal working dilutions must be determined by the end user.

Control:
Included free of charge with the antibody is 40 μg of control antigen (lyophilized powder). Reconstitute with 100 μL of deionized water. For negative control, preincubate 1 μg of purified peptide with 1 μg of antibody for one hour at room temperature. Optimal concentrations must be determined by the end user.
Presentation:
Purified immunoglobulin. Lyophilized from PBS, pH 7.4, containing 0.025% sodium azide. Reconstitute with 50 μL of sterile distilled water. Centrifuge antibody preparation before use (10,000 x g for 5 min).
Storage Conditions:
Maintain lyophilized material at -20°C for up to 6 months. After reconstitution maintain at -20°C in undiluted aliquots for up to 6 months. Avoid repeated freeze/thaw cycles.

Entrez Gene Summary:
Aquaporins are a family of small integral membrane proteins related to the major intrinsic protein (MIP or AQP0). This gene encodes an aquaporin which functions as a molecular water channel protein. It is a homotetramer with 6 bilayer spanning domains and N-glycosylation sites. The protein physically resembles channel proteins and is abundant in erythrocytes and renal tubes. The gene encoding this aquaporin is a possible candidate for disorders involving imbalance in ocular fluid movement.
UniProt Summary:
FUNCTION: SwissProt: P29972 # Forms a water-specific channel that provides the plasma membranes of red cells and kidney proximal tubules with high permeability to water, thereby permitting water to move in the direction of an osmotic gradient.
SIZE: 269 amino acids; 28526 Da
SUBUNIT: Homotetramer.
SUBCELLULAR LOCATION: Membrane; Multi-pass membrane protein.
TISSUE SPECIFICITY: Expressed in a number of tissues including erythrocytes, renal tubules, retinal pigment epithelium, heart, lung, skeletal muscle, kidney and pancreas. Weakly expressed in brain, placenta and liver.
DOMAIN: SwissProt: P29972 Aquaporins contain two tandem repeats each containing three membrane-spanning domains and a pore-forming loop with the signature motif Asn-Pro-Ala (NPA).
SIMILARITY: Belongs to the MIP/aquaporin (TC 1.A.8) family.
MISCELLANEOUS: Pharmacologically inhibited by submillimolar concentrations of mercury.
