Product Overview:
Affinity purified goat anti mouse IgG, polymeric HRP linked antibody.
Background Information:
Millipore's IHC Amplification reagent utilizes a novel controlled polymerization technology to prepare polymeric AP-linker antibody conjugates. The reagent is derived from small, multifunctional, polymerizable linkers that are used to activate a mixture of enzymes and linker antibodies, with polymerization under controlled conditions. Although the end product is a heterogeneous conjugate, with different molecular sizes, shapes and enzyme/antibody ratios, all results suggest that knowledge of the those parameters seems not to be critical with regard to performance. Compared to conventional biotin-streptavidin based three-step kits, the IHC Amplification reagent has the advantage of simpler and faster protocols, higher amplification power, biotin free and more consistent immunostaining outcomes on a variety of tissue types and antibodies (Shi et al. 1999).

PROTOCOL:
Immunohistochemistry: (All procedures should be performed at room temperature)
· Deparaffinization, rehydration
· Block endogenous peroxidase with 3% H2O2 in dd water for 10 minutes.
· Perform antigen retrieval or enzyme digestion if necessary.
1. Block with blocking solution, 10 minutes. Blot off blocking solution. No need to wash.
2. Incubate the mouse primary antibody for 30-60 minutes, wash with PBS or TBS for 5 minutes, two times. (Incubation times with the primary antibody will need to be determined by the end user.)
3. Incubate with the ready-to-use Poly-HRP-Anti-Mouse IgG for 30 minutes, wash with PBS or TBS for 5 minutes, two times.
4. Incubate with DAB or AEC solution for 5 minutes, wash with dd water.
5. Counterstain with hematoxylin and mount.
NOTES:
1. The system is biotin free so that blocking endogenous biotin is not necessary.
2. A pre-primary antibody blocking with 10% normal goat serum is recommended (CHEMICON catalog number S26-100ML).
