Anti-CD55, clone 143-30, Phycoerythrin conjugated

Recommended Replacement for: CBL512F
Species Reactivity Key Applications Host Format Antibody Type
H FC Mouse Phycoerythrin Monoclonal Antibody
Description:
Anti-CD55, clone 143-30, Phycoerythrin conjugated
Replaces:
CBL512F
Trade Name:
Chemicon (Millipore)
Specificity:
This antibody is the complement regulatory protein decay accelerating factor (DAF). It is a 70 kDa glycoprotein (in erythrocytes) anchored in the membrane by a glycosylphosphatidylinositol tail. In other cells the apparent molecular weight is somewhat larger. It has a substantial content of O-glycans, and also one N-glycan. DAF binds to activated C4b or C3b complement fragments on the cell surface, preventing the assembly and accelerating the decay of both classical and alternative pathways. DAF has a wide distribution on cells in non-haemopoietic tissues, particularly epithelium and endothelium and is specifically found at the foetal-maternal interface in placenta. Soluble forms of DAF are found, for example, in plasma, saliva, urine etc.

FUSION PARTNER: NS1 myeloma cell line
Immunogen:
PHA activated peripheral blood mononuclear cells
Clone:
143-30
Isotype:
Species Reactivity:
Human
Application Notes:
Flow Cytometry

CD55 has reduced expression on cells from individuals with paroxysmal nocturnal haemoglobinuria

Individuals with the rare Inab phenotype have an inherited deficiency of CD55

Optimal working dilutions must be determined by the end user.
Presentation:
The conjugate is supplied as a 100 test vial in phosphate buffered saline containing 10mM sodium azide and 1mg/ml bovine serum albumin. For flow cytometry we recommend using 10μL of conjugate per test.
Storage Conditions:
Store at +4°C protected from light. DO NOT FREEZE. For long term use and storage aliquot conjugate into small volumes and store at +4°C.

Unless otherwise stated in our catalog or other company documentation accompanying the product(s), our products are intended for research use only and are not to be used for any other purpose, which includes but is not limited to, unauthorized commercial uses, in vitro diagnostic uses, ex vivo or in vivo therapeutic uses or any type of consumption or application to humans or animals.
Entrez Gene Summary:
This gene encodes a protein involved in the regulation of the complement cascade. The encoded glycoprotein is also known as the decay-accelerating factor (DAF); binding of DAF to complement proteins accelerates their decay, disrupting the cascade and preventing damage to host cells. Antigens present on the DAF glycoprotein constitute the Cromer blood group system (CROM). Two alternatively spliced transcripts encoding different proteins have been identified. The predominant transcript encodes a membrane-bound protein expressed on cells exposed to plasma component proteins but an alternatively spliced transcript produces a soluble protein present at much lower levels. Additional, alternatively spliced transcript variants have been described, but their biological validity has not been determined.
Key Applications:
Flow Cytometry
UniProt Summary:
FUNCTION: SwissProt: P08174 # Also acts as the receptor for echovirus 7 and related viruses (echoviruses 13, 21, 29 and 33).
SIZE: 381 amino acids; 41400 Da
SUBUNIT: Monomer (major form) and non-disulfide-linked, covalent homodimer (minor form).
SUBCELLULAR LOCATION: Isoform 2: Cell membrane; Lipid-anchor, GPI- anchor.
TISSUE SPECIFICITY: Expressed on the plasma membranes of all cell types that are in intimate contact with plasma complement proteins. It is also found on the surfaces of epithelial cells lining extracellular compartments, and variants of the molecule are present in body fluids and in extracellular matrix.
DOMAIN: SwissProt: P08174 The first Sushi domain (SCR1) is not necessary for function. SCR2 and SCR4 provide the proper conformation for the active site on SCR3 (By similarity).
PTM: The Ser/Thr-rich domain is heavily O-glycosylated.
SIMILARITY: Belongs to the receptors of complement activation (RCA) family. & Contains 4 Sushi (CCP/SCR) domains.
