Ready to use tissue protein extract from human liver. Total cellular proteins were isolated from human liver tissue (Laemmli, 1970; Harlow & lane, 1988). Note: Source material has been tested and found to be free from HIV and Hepatitis by PCR.

SAMPLE PREPARATION: Gel Electrophoresis
Thaw the lysate at room temperature. Heat the sample at 75°C -95°C for 5 minutes. Cool on ice.Aliquot desired amount of the protein for loading on SDS-PAGE gels. The amount of protein loaded should be optimized. Should the sample need to be diluted further use 2x Sample Buffer (Laemmli, UK). Dilute 1:1 with 2x Sample Buffer.

Protein concentration from the sample was measured by taking absorbency at 280 nm using BSA. Sample was run on a 4-20% polyacrylamide gel. After electrophoresis the gel was stained with coomassie blue and examined for sharp, well resolved protein bands. There was no smeariness indicating no protein degradation. The protein was transferred to PVDF membrane and further tested for the separation and integrity of proteins by using an actin antibody.
