Suggested Procedure for Coating Cell Cultureware

1.Determine the amount of HFN needed to coat culture vessels by multiplying the total surface area (cm2) by the desired concentration (μg/mL) of HFN. Recommended amount is 2-10 μg/cm2.

2.Wet the surface of each culture vessel to be coated with a minimum amount of sterile balanced salt solution (serum and protein free) required to cover the entire area.

3.Introduce the proper CO2 atmosphere, if required.

4.Add the calculated amount of HFN to each culture vessel.

5.Allow HFN to adsorb to the surface of the vessel for 5-20 minutes.

6.Remove residual balanced salt solution before proceeding with standard cell culture procedures

The gene has three regions subject to alternative splicing, with the potential to produce 20 different transcript variants. However, the full-length nature of some variants has not been determined.

Protein/Isoform Description:
Fibronectin (FN) exists in 3 forms: plasma FN, a soluble dimeric form that becomes incororated into the Fibrin meshwork of clots; cell-surface FN, oligomers transiently attached to the cell surface; and matrix FN, highly insoluble fibrils in the extracellular matrix. The 110 kDa cell-binding fragment of FN serves as a substrate for cells that express the α5β1 Integrin receptor and other Integrin receptors that recognize the Arg-Gly-Asp (RGD) sequence. It does not contain the Hep-2/CS1 region that is recognized by α4β1 Integrin.
