Specificity:
Detects a human specific cell surface carbohydrate antigen (MW 165-170 kD) on all endothelial and selected epithelial cells. MAB430 reacts with epithelial cells in the breast salivary gland, pancreas and glomerulus. No reaction to epithelial cells in the bowel, skin and proximal tubule. The antigen recognized appears to be the sialoprotein podocalyxin protein

Background Information:
The endothelium is the layer of thin specialized epithelium, comprising a simple squamous layer of cells that line the interior surface of blood vessels, forming an interface between circulating blood in the lumen and the rest of the vessel wall (simple squamous epithelium). Endothelial cells line the entire circulatory system, from the heart to the smallest capillary.
Endothelial cells are involved in many aspects of vascular biology, such as vasoconstriction and vasodilation, and hence the control of blood pressure; blood clotting (thrombosis & fibrinolysis); atherosclerosis; formation of new blood vessels (angiogenesis) and inflammation and swelling (oedema). Endothelial cells also control the passage of materials, and the transit of white blood cells, into and out of the bloodstream.

Application Notes:
Immunohistochemistry: 1:20-1:40 (immunoperoxidase). The antigen is stable to heat and trypsin but not neuraminidase digestion. Suitable for use on tissue processed by all routine fixing and embedding techniques.

Immunohistochemistry on paraffin embedded tissues: trypsin digestion or HEIR antigen retrieval suggested.

Optimal working dilutions must be determined by end user.

This gene encodes a member of the sialomucin protein family. The encoded protein was originally identified as an important component of glomerular podocytes. Podocytes are highly differentiated epithelial cells with interdigitating foot processes covering the outer aspect of the glomerular basement membrane. Other biological activities of the encoded protein include: binding in a membrane protein complex with Na+/H+ exchanger regulatory factor to intracellular cytoskeletal elements, playing a role in hematopoetic cell differentiation, and being expressed in vascular endothelium cells and binding to L-selectin.
