Product Overview:
The Astrocyte Differentiation Medium is a specially formulated medium optimized for the preferential differentiation of rodent neural stem cells to an astrocyte lineage. The medium has been extensively validated on mouse cortical and spinal cord neural stem cells and on rat hippocampal neural stem cells. Using Millipore's Astrocyte Differentiation Medium, relatively pure populations of astrocytes (>80-90% GFAP) are obtained. Very low levels of neurons (<1% MAP2A,2B positive) or oligodendrocytes (<5% O1 positive) are detected. Sufficient medium is supplied to provide for 100 separate reactions at 1 mL volume or 10 reactions at 10 mL volume.

The Rat Astrocyte Differentiation Medium contains DMEM/F12 w/o HEPES, w/ L-Glutamine (Millipore Catalogue No. DF-042-B), 1X B27-CHEM, 2 mM L-Glutamine (Millipore Catalogue No. TMS-002-C), 1X Penicillin Streptomycin (Millipore Catalogue No. TMS-AB2-C), BMP-4, 50 ng/mL LIF (Millipore Catalogue No. ESG1107) and 1% heat-inactivated fetal bovine serum (Millipore Catalogue No. ES-009-B)
Storage Conditions:
SCM010A, Astrocyte Differentiation Medium should be stored at -20°C until ready to use. Upon thawing, the basal medium should be stored at 2-8°C and given a 1-month expiration dating.

FGF-2 (10 μg, lyophilized) should be reconstituted with 100 μL 5mM Tris-HCl, pH 7.6 for a final concentration of 100 μg/mL. Dispense into aliquots to avoid repeated freeze/thaw.
