Introduction:
Neural stem cells (NSC) are self-renewing cells with the capacity to differentiate into neurons, astrocytes and oligodendrocytes. Adult NSC are extremely rare and are believed to reside only in discrete regions of the hippocampus and the subventricular zone. Due to their therapeutic promise, considerable attention has been focused on identifying the sources of stem cells, the signals that regulate their proliferation and the specification of neural stem cells towards more differentiated cell lineages (1).
Presently, neural stem cells are often identified based upon the presence of molecular markers that are correlated with the stem and/or progenitor state along with the absence of differentiated phenotypes as assessed through marker analysis. To aid researchers in the accurate identification and characterization of neural stem cells, Millipore presents the Adult Rat Hippocampal Neural Stem Cell Kit (Millipore Catalog No. SCR021).
Millipore's Adult Rat Hippocampal Neural Stem Cell Kit provides ready-to use primary neural stem cells isolated from the hippocampus of adult Fisher 344 rats along with antibodies for the characterization of the neural stem cell population (Nestin and Sox 2) and their differentiated progenies (MAP-2 for neurons and GFAP for astrocytes). Mouse and rabbit immunoglobulins for the assessment of background staining are also included.
All of the antibodies provided in the kit have been tested and optimized for use in immunocytochemistry on adult rat neural stem cells. We recommend that Millipore's Adult Rat Hippocampal Neural Stem Cell Kit (Millipore Catalog No. SCR021 or SCR022) be used in conjunction with differentiation assays that demonstrate multipotentiality of the starting cell population. Please look for these differentiation assays from Millipore in the near future.

- Rabbit anti-Sox 2: (Part No. 2003600) One vial containing 20 μg affinity purified polyclonal antibody. Store at 2° to 8°C.

When stored at the recommended storage conditions (refer to Kit Components), components are stable up to the expiration date. Do not expose to elevated temperatures. Discard any remaining reagents after the expiration date. We recommend that the cells be used within ten passages.
