Product Overview:
The Cardiac Stem Cell Isolation Kit provides enough reagents for five separate isolations. Included in the kit are (1) ready to use isolation and dissociation buffers for the surgical resections of the heart, (2) five individually packaged, self-contained Steriflip® filtration devices for the efficient and sterile separation of dissociated cells from ventricular tissues, (3) two differential gradient solutions for the rapid purification of Sca-1 positive CSCs, (4) a 500 mL bottle of Cardiac Stem Cell Maintenance Medium for the expansion of acutely isolated CSCs, and (5) a 500 mL bottle of Cardiomyocyte Differentiation Medium for the preferential differentiation of isolated CSCs to cardiomyocytes.
Background Information:
The isolation and expansion of cardiac stem cells (CSCs) opens new opportunities to the field of cardiac regenerative medicine. CSCs have recently been isolated from human and murine tissues based upon cell biomarkers (Sca-1, TERT, c-Kit, side population), demonstrating the presence of a non-circulating stem cell niche within the myocardium that is estimated to account for 11-14% of the total cell population. These cells appear to be bi-potent in their capacity to form cardiomyocytes and vascular endothelial cells. Moreover, preliminary engraftment studies suggest that these cells are ideal candidates for future research on cardiac regeneration. However, the rarity of CSCs coupled with the complex isolation procedure pose a significant challenge for the advancement of the field. To overcome this obstacle, Millipore has developed an easy-to-use cell isolation kit that is capable of obtaining a high-yield, pure population of CSCs. This advancement allows investigators the opportunity to obtain a significantly greater number of CSCs for their studies without the need for time consuming, complex protocols and expensive cell sorting equipment.

Storage Conditions:
Cardiac Stem Cell Isolation Buffer: The buffer is provided ready to use. Store and maintain buffer at 2-8°C in the dark. When stored as recommended, the buffer is stable for up to 1 year. Buffer must be maintained in sterile conditions as contamination can seriously compromise stability.

Cardiac Tissue Dissociation Buffer: Prior to initial use, thaw frozen buffer at 37°C until it has been completely equilibrated. Maintain thawed solution at 2-8°C in the dark. When stored under appropriate conditions, the buffer is stable for up to one year.

Steriflip 100 mm Nylon Net NY1H RS: When stored at room temperature, the Steriflip units are stable for up to 2 years.

Gradient Solutions 1 & 2: Store and maintain buffer at 2-8°C in the dark. Before use, mix the contents of bottle thoroughly. When stored at 2-8°C and maintained under sterile conditions, the gradient solutions are stable for up to one year. At -20°C, the solutions can only be stored approximately six months.

Cardiac Stem Cell Maintenance Medium and Cardiomyocyte Differentiation Medium: For long term storage, maintain the media at -20°C. Prior to initial use, thaw frozen media at 4°C overnight or until it has become completely equilibrated. Maintain thawed media at 2-8°C in the dark. For Differentiation Medium, warm an appropriate aliquot of media to be used the day of at 37°C. The medium comes ready to use. When stored under appropriate conditions medium is stable for up to one year.
