Anti-α-Catenin
Background Information:
The catenins are polypeptides that bind to the conserved cytoplasmic tail of cadherins and are required for cadherin function (Gumbiner et al. 1993). a-catenin is a 102 kDa protein that is associated with the cadherin indirectly through its interaction with beta-catenin and gamma-catenin (Fouquet et al. 1992). beta-Catenin and its homolog plakoglobin associate directly with the cadherin (Butz et al. 1994). Thus, the cell-cell adherens junction is a structure composed of the transmembrane cadherin, which is associated directly with either beta-catenin or plakoglobin, which in turn assoctiates directly with alpha-catenin. alpha-catenin then mediates the interaction between the cadherin-catenin complex and the actin cytoskeleton (Nagafuchi et al. 1994; Shahidul et al. 1996). alpha-catenin has been characterized as a tumor suppressor, suggesting that it may have a regulatory function in regulation to its structural role (Rudiger 1998).
