Background Information:
Catenins are cytosolic proteins found in varying abundance in many developing and adult tissues. Plakoglobin and beta-catenin (95 kDa) are homologous proteins and can subsitute for one another in the cadherin/catenin complex. Their activities are controlled by three types of interactions: those with cadherins in adherens junctions, linking them to the actin cytoskeleton; interactions in the nucleus, where they bind to transcription factors and stimulate gene expression; interactions of free cytoplasmic beta-catenin with axin and adenomatous polyposis coli (APC) protein which target it for degradation. Studies in the past year have demonstrated the complex interplay between these three types of interactions and the different behavior of beta-catenin and plakoglobin. Their involvement in morphogenesis and tumorigenesis strongly suggests that catenins play key roles in adhesion-mediated signaling.

Immunocytochemistry:
A previous lot of this antibody was used in immunocytochemistry.

Immunoprecipitation:
A previous lot of this antibody was used in immunoprecipitation.

Immunohistochemistry(paraffin):
A 1:250-1:500 dilution from a previous lot was used on formalin-fixed paraffin-embedded tissues*.
*Requires pressure cooker antigen retrieval method. Sections are baked overnight at 60°C, and then deparaffinized and treated for antigen retrieval by immersion in 6.6mM sodium citrate pH 6.0 for 5 minutes in a conventional pressure cooker. Sections were then blocked with 0.3% bovine serum albumin in Tris-buffered saline pH 8.0. Incubations are usually overnight, before washing 7 times with TBS plus 0.01% triton X-100 in the sixth wash.

ELISA:
A previous lot of this antibody was used in ELISA.
Western blot Analysis:1:500 dilution of this lot was used.
Optimal working dilutions must be determined by end user.

Handling Recommendations:
Upon receipt, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Beta-catenin is an adherens junction protein. Adherens junctions (AJs; also called the zonula adherens) are critical for the establishment and maintenance of epithelial layers, such as those lining organ surfaces. AJs mediate adhesion between cells, communicate a signal that neighboring cells are present, and anchor the actin cytoskeleton. In serving these roles, AJs regulate normal cell growth and behavior. At several stages of embryogenesis, wound healing, and tumor cell metastasis, cells form and leave epithelia. This process, which involves the disruption and reestablishment of epithelial cell-cell contacts, may be regulated by the disassembly and assembly of AJs. AJs may also function in the transmission of the 'contact inhibition' signal, which instructs cells to stop dividing once an epithelial sheet is complete.[supplied by OMIM].

The other pool is part of a large complex containing AXIN1, AXIN2, APC, CSNK1A1 and GSK3B that promotes phosphorylation on N-terminal Ser and Thr residues and ubiquitination of CTNNB1 via BTRC and its subsequent degradation by the proteasome. Wnt- dependent activation of DVL antagonizes the action of GSK3B. When GSK3B activity is inhibited the complex dissociates, CTNNB1 is dephosphorylated and is no longer targeted for destruction. The stabilized protein translocates to the nucleus, where it binds TCF/LEF-1 family members, TBP, BCL9 and possibly also RUVBL1 and DUPLIN. Binds CTNNBIP and EP300. CTNNB1 forms a ternary complex with LEF1 and EP300 that is disrupted by CTNNBIP1 binding (By similarity). SUBCELLULAR LOCATION: Cytoplasm. Nucleus. Note=Cytoplasmic when it is unstabilized (high level of phosphorylation) or bound to CDH1. Translocates to the nucleus when it is stabilized (low level of phosphorylation). Interaction with GLIS2 promotes nuclear translocation.
TISSUE SPECIFICITY: Expressed in several hair follicle cell types: basal and peripheral matrix cells, and cells of the outer and inner root sheats. Expressed in colon.
