Anti-Aquaporin 4, C-terminus

Species Reactivity Key Applications Host Format Antibody Type
H, M, R IH, WB Rabbit Affinity Purified Polyclonal Antibody
Description:
Anti-Aquaporin 4, C-terminus
Trade Name:
Chemicon (Millipore)
Specificity:
Recognizes aquaporin 4. On western blots of rat brain membranes, the antibody detects a band migrating at approximately 38 kDa corresponding to aquaporin 4.
Molecular Weight:
38 kDa
Epitope:
C-terminus
Immunogen:
GST fusion protein with residues 249-323 of rat Aquaporin 4 (SwissProt accession number P47863). It is expected that the antibody may also react with mouse (73/75), bovine (71/75), human (69/75) and rabbit (64/75) due to sequence homology.
Background Information:
Water is a critical component of all living cells. Interestingly, tissue membranes show a great degree of water permeability. Mammalian red cells, renal proximal tubules, and descending thin limb of Henle are extraordinarily permeable to water. Water crosses hydrophobic plasma membranes either by simple diffusion or through a facilitative transport mechanism mediated by special proteins "aquaporins". Aquaporins, are integral membrane proteins from a larger family of major intrinsic proteins (MIP) that form pores in the membrane of biological cells. Over the last decade, genes for several members of aquaporin family have been cloned, expressed, and their distribution studied in many tissues. The water-specific channel Aquaporin 4 is located in various tissues including the gastrointestinal tract, kidney and brain. It is an osmoreceptor which regulates body water balance and mediates water flow within the central nervous system. In CNS tissue, AQP4 localizes to astrocyte endfeet making it a useful structural marker.
Species Reactivity:
  • Human
  • Mouse
  • Rat
Application Notes:
Western blotting: 1:500 on rat brain membranes.

Immunohistochemistry

Dilutions should be made using a carrier protein such as BSA (1-3%).

Optimal working dilutions must be determined by the end user.
Control:
CONTROL ANTIGEN: Included free of charge with the antibody is 120 μg of control antigen (lyophilized powder). Reconstitute with 100 μL of PBS. For positive control, in Western blot using 20 ng of protein per Minigel lane. For negative control, preincubate 3 μg of purified peptide with 1 μg of antibody for one hour at room temperature. Optimal concentrations must be determined by the end user.
Purification Method:
Immunoaffinity purified
Presentation:
Affinity Purified immunoglobulin. Lyophilized from PBS, pH 7.4, containing 1% BSA, and 0.025% sodium azide. Reconstitute with 50 μL of sterile distilled water. Centrifuge antibody preparation before use (10,000 x g for 5 min).
Storage Conditions:
Maintain lyophilized material at -20°C for up to 6 months. After reconstitution maintain at -20°C in undiluted aliquots for up to 6 months. Avoid repeated freeze/thaw cycles.

Entrez Gene Summary:
This gene encodes a member of the aquaporin family of intrinsic membrane proteins that function as water-selective channels in the plasma membranes of many cells. The encoded protein is the predominant aquaporin found in brain. Two alternatively spliced transcript variants encoding distinct isoforms have been found for this gene.
