PrecisION hNav1.3-CHO Recombinant Cell Line

Description:
PrecisION hNav1.3-CHO Recombinant Cell Line
Trade Name:
Upstate (Millipore)
Qty/Pk:
Product Overview:
Recombinant CHO-K1 cell line expressing the human Nav1.3 sodium ion channel
Background Information:
Nav1.3 is a voltage gated sodium channel which is expressed during fetal development and adult patho-physiolgical conditions. For example, as well as being normally expressed in embryonic dorsal root ganglia, it is also up regulated in damaged adult peripheral nerves and associated with increased electrical excitability in neuropathic pain states.
Application Notes:
CHO-K1 cells stably expressing hNav1.3 were analysed by patch-clamp techniques giving typical peal currents range between 0.8-6 nA. With IonWorks, typically greater than 90% of cells expressed peak currents of at least 2 nA. IC50 values obtained for the known hNav1.3 inhibitors were 4 nM for tetrodotoxin (patch clamp) and 32 mM for tetracaine (VIPR).
Usage Statement:
Unless otherwise stated in our catalog or other company documentation accompanying the product(s), our products are intended for research use only and are not to be used for any other purpose, which includes but is not limited to, unauthorized commercial uses, in vitro diagnostic uses, ex vivo or in vivo therapeutic uses or any type of consumption or application to humans or animals.
Entrez Gene Summary:
Voltage-gated sodium channels are transmembrane glycoprotein complexes composed of a large alpha subunit with 24 transmembrane domains and one or more regulatory beta subunits. They are responsible for the generation and propagation of action potentials in neurons and muscle. This gene encodes one member of the sodium channel alpha subunit gene family, and is found in a cluster of five alpha subunit genes on chromosome 2. Multiple transcript variants encoding different isoforms have been found for this gene.
UniProt Summary:
FUNCTION: SwissProt: Q9NY46 # Mediates the voltage-dependent sodium ion permeability of excitable membranes. Assuming opened or closed conformations in response to the voltage difference across the membrane, the protein forms a sodium-selective channel through which Na(+) ions may pass in accordance with their electrochemical gradient.
SIZE: 2000 amino acids; 226294 Da
SUBUNIT: The sodium channel consists of a large polypeptide and 2- 3 smaller ones. This sequence represents a large polypeptide. Interacts with NEDD4L.
SUBCELLULAR LOCATION: Membrane; Multi-pass membrane protein.
DOMAIN: SwissProt: Q9NY46 The sequence contains 4 internal repeats, each with 5 hydrophobic segments (S1,S2,S3,S5,S6) and one positively charged segment (S4). Segments S4 are probably the voltage-sensors and are characterized by a series of positively charged amino acids at every third position.
PTM: May be ubiquitinated by NEDD4L; which would promote its endocytosis.
SIMILARITY: Belongs to the sodium channel family. & Contains 1 IQ domain.
