Recombinant HEK293 cell line expressing the human Nav1.6 (type VIII voltage-gated sodium channel alpha subunit).

Format:
2 x 1 ml aliquots containing 12.22 x 106cells/ml in 10% DMSO at passage 11.
Background Information:
hNav1.6 is primarily expressed in the output neurones of the cerebellum, cortex and hippocampus of the CNS and in the Nodes of Ranvier of sensory and motor axons of the PNS. hNav1.6 is involved with the initiation and transmission of action potentials in central neurons and their myelinated axons. Mutation or disruption of the Nav1.6 gene causes cerebellar ataxia and motor end plate disease in mice. This channel is a potential target for analgesic and antiepileptic drugs.
Application Notes:
HEK293 cells stably expressing hNav1.6 were analyzed by patch-clamp and IonWorks™. Typically, >90% of cells sealed with >80% of cells expressing giving mean peak currents of 1.42 nA (n=313). IC50 values obtained for the known hNav1.6 inhibitors were 2.5 nM for tetrodotoxin and for lamotrigine (Lamictal) were 486 μM, 127 μM, and 31 μM at VH of -120 mV, -90 mV & -60 mV respectively.
