Background Information:
Kv1.2 frequently forms heteromultimers with other Kv1.x subunits contributing to the delayed rectifier potassium current in a variety of tissues. The channel is expressed throughout the brain, spinal cord, Schwann cells , the heart atria and ventricles, pancreatic islets, retina and smooth muscle. It is primarily involved with maintaining the resting potential of cells and modulating electrical excitability in neurons and muscles
Application Notes:
CHO-K1 cells expressing hKv1.2 were characterized in terms of their pharmacological and biophysical properties using whole-cell patch clamp techniques. The mean current amplitude at 40 mV was 2000 &plusms; 570 pA (n=6). The threshold of current activation was around -10 mV and exhibited slow C-type inactivation at voltages >/= 40 mV. The current activates very slowly between -10 mV and 20 mV. These electrophysiological properties are consistent with previous reports in the literature describing Kv1.2 currents. The cell-line was shown to be selectively expressing hKv1.2 current since it was blocked by low nM concentrations of the toxin blocker, Tityustoxin Kα and resistant to low mM concentrations of TEA. Functional channel expression over time was monitored using IonWorks™ HT. Channel expression is robust over at least 32 passages: 91% of cells expressed Kv1.2 currents (>500 pA) at passage 32 giving a mean current amplitude of 1390 ± 69 pA at 60 mV (n=114).

Their diverse functions include regulating neurotransmitter release, heart rate, insulin secretion, neuronal excitability, epithelial electrolyte transport, smooth muscle contraction, and cell volume. Four sequence-related potassium channel genes - shaker, shaw, shab, and shal - have been identified in Drosophila, and each has been shown to have human homolog(s). This gene encodes a member of the potassium channel, voltage-gated, shaker-related subfamily. This member contains six membrane-spanning domains with a shaker-type repeat in the fourth segment. It belongs to the delayed rectifier class, members of which allow nerve cells to efficiently repolarize following an action potential. The coding region of this gene is intronless, and the gene is clustered with genes KCNA3 and KCNA10 on chromosome 1.
