Background Information:
Kv1.4 is a voltage-gated, fast-inactivating, A-type potassium ion channel also known as HuK(II), hPCN2, HK1, RCK4, RHK1, RK4, RK8 and MK4. It is expressed in the CNS, lung, skeletal muscle, heart and pancreatic islet cells. It is responsibly for neuronal afterhypolarization. The expression of Kv1.4 has been shown to increase after induction of diabetes and in ventricular myocytes after a heart attack
Application Notes:
CHO-K1 cells expressing hKv1.4 were characterised in terms of their pharmacological and biophysical properties using whole-cell patch clamp techniques. The mean current amplitude at 0 mV was 3600 ± 2700 pA (n=3). The threshold of activation was around -40 mV and exhibited fast N-type inactivation. The kinetics of inactivation and the current/voltage relationship are consistent with published data describing the Kv1.4 current. The current was blocked by 1 mM 4-AP to an extent expected for selective expression of Kv1.4 channels. The percent inhibition of the current using 1 mM 4-AP was 71 ± 6% (n=4). The current was also inhibited by mild acidosis in agreement with previous reports describing the Kv1.4 current. Functional channel expression over time was monitored using IonWorks™ HT. Channel expression is robust over at least 36 passages: 95% of cells expressed Kv1.4 currents at passage 35 giving a mean current amplitude of 4.64 ± 0.22 nA at 60 mV (n=141).

Their diverse functions include regulating neurotransmitter release, heart rate, insulin secretion, neuronal excitability, epithelial electrolyte transport, smooth muscle contraction, and cell volume. Four sequence-related potassium channel genes - shaker, shaw, shab, and shal - have been identified in Drosophila, and each has been shown to have human homolog(s). This gene encodes a member of the potassium channel, voltage-gated, shaker-related subfamily. This member contains six membrane-spanning domains with a shaker-type repeat in the fourth segment. It belongs to the A-type potassium current class, the members of which may be important in the regulation of the fast repolarizing phase of action potentials in heart and thus may influnce the duration of cardiac action potential. The coding region of this gene is intronless, and the gene is clustered with genes KCNA3 and KCNA10 on chromosome 1.
