Recombinant CHO-K1 cell line expressing the human voltage-gated potassium channel Kv1.7.
Background Information:
Kv1.7 is a voltage-gated, delayed rectifier, potassium channel. It is expressed in the heart, skeletal muscle and pulmonary arteries. As this channel has properties similar to the ultra-rapidly activating (IKur) current of the heart, it is potentially a candidate channel for involvement in human cardiac disorders.
Application Notes:
CHO-K1 cells expressing hKv1.7 were characterized in terms of their pharmacological and biophysical properties using whole-cell patch clamp techniques. The mean current amplitude at 60 mV was 2.69 ± 0.8 nA (n=4). The threshold for current activation was around -20 mV and exhibited a very slow C-type inactivation at depolarized potentials. TEA (50 mM) significantly blocked the current (56-61%) which is consistent with previous reports describing Kv1.7 sensitivity to TEA. 4-AP (100 μM) also blocked the current (82%) as expected for the selective expression of Kv1.7 channels. Similarly ShK (10 mM/50 mM) blocked the current by 17.5%/30.6% respectively. Functional expression over time was monitored using IonWorks™ HT. Channel expression is robust over at least 44 passages: 94.2% of cells expressed Kv1.7 currents of >500 pA at passage 44.

This gene encodes a member of the potassium channel, voltage-gated, shaker-related subfamily. This member contains six membrane-spanning domains with a shaker-type repeat in the fourth segment. The gene is expressed preferentially in skeletal muscle, heart and kidney. It is a candidate gene for inherited cardiac disorders.
