PrecisION hKv3.2-CHO Recombinant Cell Line

Description:
PrecisION hKv3.2-CHO Recombinant Cell Line
Trade Name:
Upstate (Millipore)
Qty/Pk:
Product Overview:
Recombinant CHO-K1 cell line expressing the human potassium channel Kv3.2.

Format: 2 x 1 ml aliquots containing 2.64 x 106 cells/ml at passage 7.
Background Information:
Kv3.2 channels are expressed in neurons known to fire at high frequency. Their biophysical properties could enable cells to generate the fast chains of action potentials involved in neuronal excitability. They are thought to play a role in the regulation of the human 24hr circadian rhythm, as they are highly expressed in the suprachiasmatic nucleus. Kv3.2 channels are also found in the insulin secreting beta-cells of the pancreas and their modulation could provide novel targets in the treatment of type-2 diabetes.
Application Notes:
CHO-K1 cells expressing hKv3.2 were characterised in terms of their pharmacological and biophysical properties using whole-cell patch clamp techniques and IonWorksTM HT. Using whole-cell patch clamp techniques the threshold of current activation was found to occur when the membrane potential was depolarised to potentials more positive than -30 mV. The mean current at +40 mV was 9.55 ± 4.10 nA (n=7). Currents were inhibited by the known potassium channel blockers TEA and 4-AP in sub and low mM concentrations. The currents obtained with IonWorksTM HT were typical of homomeric hKv3.2 currents since they had an initial fast transient component followed by a sustained component. Functional channel expression over time was monitored using IonWorksTM HT. Channel expression is robust over at least 30 passages. At passage 30, 88% of cells sealed with a resistance greater than 50 MOhm. Of these, 100% expressed hKv3.2 currents greater than 500 pA with a mean current amplitude of 6.80 ± 0.12 nA (n=169).

Entrez Gene Summary:
The Shaker gene family of Drosophila encodes components of voltage-gated potassium channels and is comprised of four subfamilies. Based on sequence similarity, this gene is similar to one of these subfamilies, namely the Shaw subfamily. The protein encoded by this gene belongs to the delayed rectifier class of channel proteins and is an integral membrane protein that mediates the voltage-dependent potassium ion permeability of excitable membranes. Three transcript variants encoding three different isoforms have been found for this gene.
UniProt Summary:
FUNCTION: SwissProt: Q96PR1 # Mediates the voltage-dependent potassium ion permeability of excitable membranes. Assuming opened or closed conformations in response to the voltage difference across the membrane, the protein forms a potassium-selective channel through which potassium ions may pass in accordance with their electrochemical gradient. Channel properties are modulated by subunit assembly (By similarity).
SIZE: 638 amino acids; 70226 Da
SUBUNIT: The potassium channel is probably composed of a homo- or heterotetrameric complex of pore-forming subunits that can associate with modulating accessory subunits. Interacts with KCNC1, KCNE1, KCNE2 and KCNE3 (By similarity).
SUBCELLULAR LOCATION: Membrane; Multi-pass membrane protein.
DOMAIN: SwissProt: Q96PR1 The segment S4 is probably the voltage-sensor and is characterized by a series of positively charged amino acids at every third position. & The tail may be important in modulation of channel activity and/or targeting of the channel to specific subcellular compartments.
SIMILARITY: Belongs to the potassium channel family. C (Shaw) subfamily.
