Product Overview:
The FlowCellect T CD4 Cell FAS kit allows for the identification and counts of CD3 T Cells and CD4 T helper cells and the level of FAS expression levels enabeling for the detection and quantitation of CD95 (Apo1, FAS) marker on either whole blood or PBMC using simplified no-wash assays. The kit can be utilized on any flow cytometry system equipped with blue laser. The performance of these assays on the guava easyCyte 8HT platform along with the Incyte software allows for cell count information on CD4, CD3 T cells and count and % of populations expressing CD95/FAS. Furthermore, easy identification of activated T cells can be performed using heat map features or measurement of the kinetics of activation using the dose response features. The simplified identification of activated samples can be of great utility in drug development, kinetic studies and understanding mechanism of compound action and disease.
Background Information:
FAS expression and apoptosis has been shown to play a significant role in T-cell selection in the thymus for the acquisition of self-tolerance, and in T-cell mediated cytotoxicity and in downregulation of the immune response. It has also a role in the clonal deletion of activated T-cells in order to maintain T cell homeostasis, down-regulation of inflammatory T cells and the regulation of activated B cells. The presence or absence of FAS and/or FasL on cells of the immune system and their interaction with FAS and FasL on cells of other tissues plays a major factor in immune system homeostasis. It has been proposed that AICD serves a role in limiting the expansion of an immune response by eliminating lymphocytes that are no longer necessary at the end of an infection. Recent studies have also indicated that in addition to a critical role in receptor mediated apoptosis, FAS has a growth-promoting role during tumorigenesis.
