Specificity:
Tyrosine Hydroxylase. Recognizes a protein of approximately 59-63 kDa by Western blot. Does not react with the following on Western Blots: dopamine-beta-hydroxylase, phenylalanine hydroxylase, trytophan hydroxylase, dehydropteridine reductase, sepiapterin reductase, phenethanolamine-N-methyl transferase (PNMT).

Recognizes an epitope on the outside of the regulatory N-terminus of tyrosine hydroxylase.

Background Information:
Tyrosine hydroxylase plays an important role in the physiology of adrenergic neurons. It is the first and rate-limiting enzyme involved in the biosynthesis of the catecholamines Dopamine and Norepinephrine from tyrosine. TH is, therefore, a useful marker for dopaminergic and noradrenergic neurons. The enzymatic activity of TH requires ferrous ions as cofactors and is believed to be regulated by phosphorylation. At least four isoforms of human TH have been identified which result from alternative splicing.

Application Notes:
Antibody is prediluted and ready to use for Immunohistochemistry of formalin-fixed, paraffin-embedded tissues.

Pretreatment: Heat Induced Epitope Retrieval (HIER). Recommend Citrate Buffer, pH 6.0 (Cat. No. 21545). No signal was detected without Epitope retrieval.

Incubation: 30 minutes with IHC Select® Detection Kits.

Tyrosine Hydroxylase has been prediluted for use as the primary antibody with Chemicon's IHC Select® Detection Kits and Protocols (Catalog Nos. DAB050, DET-HP1000, APR050, and DET-APR1000), but other supplier's IHC detection systems may be used. For optimized protocol details, visit www.chemicon.com and select the protocols link under Cat.
