Purity: The antibody is shown to be >95% pure as determined by SDS-PAGE. The BimS antibody is supplied 0.22 micron filtered.
Specificity:
Purified rat monoclonal antibody IgG2a Kappa isotype raised against recombinant mouse BimShort (BimS) protein. The antibody recognises a reactive epitope that is common to all splice variants of Bim including BimExtra Long, BimLong and BimShort. These antibodies can be used for the detection of Bim by standard techniques including Western blot, immunoprecipitation, immunohistochemistry and FACS. This antibody is highly sensitive and can, in most cases, recognize endogenous levels of BimS*.

Background Information:
Bim (bcl-2-interacting mediator of cell death) is a proapoptotic member of the Bcl-2 family that shares only the BH3 domain with this family. There are three isoforms of Bim: BimEL, BimL, and BimS. Bim is involved in regulating the intrinsic mitochondrial apoptotic pathway by inducing cytochrome c release, which in turn, activates caspase-9 and then caspase-3. Bim also plays a critical role in central and peripheral deletion of T lymphocytes and in controlling B cell homeostasis and activation. BimEL, the long isoform of Bim, is a ~28 kDa protein that is predominantly expressed in T and B cells and is activated by ERK1/2 pathway. The activation of BimEL by ERK1/2 promotes its phosphorylation on serine 65 (serine 69 in human), targeting it for degradation via the proteasome.

Application Notes:
The antibody is intended for the detection of all splice variants of Bim in a range of techniques including:

Western blot: ~17-23kDa on reduced westerns; Bim(L) ~19kDa; ~23 kDa BimEL, Bim(s) ~17kDa (very difficult to detect at normal levels) ; endogenous levels are difficult to detect without prior immunoprecipitation of Bim isoforms in most cases. See O'Reilly, 2000 for complete details.

Tissue specificity: Isoform BimEL, isoform BimL and isoform BimS are the predominant isoforms and are ubiquitously expressed with a tissue-specific variation.
