Description:
Anti-NG2 Chondroitin Sulfate Proteoglycan
Trade Name:
Chemicon (Millipore)
Specificity:
NG2 Chondroitin Sulfate Proteoglycan. MAB5384 reacts with native, recombinant and protein from cells which express NG2.
Molecular Weight:
~ 280 kDa
Immunogen:
Cell line expressing a truncated form of NG2.
Clone:
132.39
Isotype:
Background Information:
NG2 is a high molecular weight, integral membrane chondroitin sulfate proteoglycan. It is found on the surfaces of an unusual class of glial cells within the developing and mature central nervous system that have the properties of oligodendrocyte precursor cells (i.e., O-2A progenitor cells). NG2 is also found on the surfaces of chondroblasts, proliferating capillary endothelial cells, some human melanoma cell lines, and on leukemic blasts in childhood acute lymphoblastic leukemia. The NG2 proteoglycan is likely to play a role in regulation of cell motility, axon outgrowth and the cellular responses to platelet-derived growth factor.
Species Reactivity:
  • Human
  • Mouse
  • Rat
Species Reactivity Note:
Rat. Expected to react with human and mouse based on sequence similarity.
Application Notes:
Immunocytochemistry: 1:200-1:500 dilution of a previous lot was used on oligodendrocyte precursor cells.

ELISA (direct): A previous lot of this antibody was used in ELISA.

Optimal working dilutions must be determined by end user.
Control:
Neonatal rat brain extract
Quality Assurance:
Evaluated by Western Blot on rat brain lysates.

Western Blot Analysis: 1:500 dilution of this antibody detected NG2 Chondroitin Sulfate Proteoglycan on 10 µg of rat brain lysates.

Entrez Gene Summary:
extracellular matrix protein secreted by many cell types; member of the large chondroitin sulfate proteoglycan (CSPG) family; may be involved with angiogenesis -dependent processes such as wound healing or tumor growth [RGD]
UniProt Summary:
FUNCTION: Proteoglycan playing a role in cell proliferation and migration which stimulates endothelial cells motility during microvascular morphogenesis. May also inhibit neurite outgrowth and growth cone collapse during axon regeneration. Cell surface receptor for collagen alpha 2(VI) which may confer cells ability to migrate on that substrate. Binds through its extracellular N- terminus growth factors, extracellular matrix proteases modulating their activity. May regulate MPP16-dependent degradation and invasion of type I collagen participating in melanoma cells invasion properties. May modulate the plasminogen system by enhancing plasminogen activation and inhibiting angiostatin. Functions also as a signal transducing protein by binding through its cytoplasmic C-terminus scaffolding and signaling proteins. May promote retraction fiber formation and cell polarization through Rho GTPase activation. May stimulate alpha-4, beta-1 integrin- mediated adhesion and spreading by recruiting and activating a signaling cascade through CDC42, ACK1 and BCAR1. May activate FAK and ERK1/ERK2 signaling cascades.

SIZE: 2326 amino acids; 251909 Da

SUBUNIT: Interacts with GRIP1, GRIP2 and GRIA2. Forms a ternary complex with GRIP1 and GRIA2. Interacts with ITGA4 through its chondroitin sulfate glycosaminoglycan. Interacts with BCAR1, CDC42 and ACK1. Interacts with MMP16 (By similarity). Interacts with the first PDZ domain of MPDZ. Interacts with PRKCA. Interacts with LGALS3 and the integrin composed of ITGB1 and ITGA3. Binds TNC, laminin-1, COL5A1 and COL6A2. Interacts with PLG and angiostatin. Binds FGF2 and PDGFA.

SUBCELLULAR LOCATION: Cell membrane; Single-pass type I membrane protein; Extracellular side. Apical cell membrane; Single-pass type I membrane protein; Extracellular side. Cell projection › lamellipodium membrane; Single-pass type I membrane protein; Extracellular side. Note= Localizes to the plasma membrane of oligodendrocytes (By similarity). Localized at the apical plasma membrane it relocalizes to the lamellipodia of astrocytoma upon phosphorylation by PRKCA. Localizes to the retraction fibers. A fraction may undergo cell surface proteolysis and secretion.

TISSUE SPECIFICITY: Neural cells and also extraneural tissues, especially in the developing mesenchyme.

PTM: N-glycosylated (By similarity). & O-glycosylated; contains glycosaminoglycan chondroitin sulfate which are required for proper localization and function in stress fiber formation. Involved in interaction with MMP16 and ITGA4. Phosphorylation by PRKCA regulates its subcellular location and function in cell motility.

MISCELLANEOUS: Valuable marker for several incompletely differentiated precursor cells.
