At the onset of apoptosis it proteolytically cleaves poly(ADP-ribose) polymerase (PARP) at a '216-Asp- -Gly-217' bond. Cleaves and activates sterol regulatory element binding proteins (SREBPs) between the basic helix-loop- helix leucine zipper domain and the membrane attachment domain. Cleaves and activates caspase-6, -7 and -9. Involved in the cleavage of huntingtin.

Background Information:
Caspase-3/Yama/Apopain, also known as proICE and CPP32, is a member of the ICE-like protease family. Caspase-3 is expressed as a 32 kDa precursor, which is proteolytically cleaved to yield an active form comprised of 17kDa and 12kDa subunits, respectively. The active enzyme appears to play an important role in the regulation or execution of apoptosis. One of its target substrates is the enzyme poly (ADP-ribose) polymerase (PARP) which is involved in DNA repair and maintenance of genome integrity.
Specificity:
Recognizes full-length Caspase 3 (Yama/Apopain) and proteolytic fragments.
