The ChemiBlot Molecular Weight Markers is a mixture of artificial IgG-binding proteins with one to six times linearly repeated structure of domains A-B of Staphylococcal protein A [pA(AB1 to pA(AB)6] 1 The six artificial proteins are separately synthesized by genetic engineered E. coli and are purified by affinity chromatography on an IgG column. This product shows six distinct bands on SDS-PAGE. The markers can be used for estimation of molecular weight on SDS-PAGE and as a positive control for Western blot 3

MOLECULAR WEIGHTS:

14,800, 28,201, 41,603, 55,004, 68,406 and 81,807. The molecular weights are calculated from their amino acid composition.

Western Blotting

1) Estimation of molecular weight on SDS-PAGE: 10 μL/lane for CBBR (Coomassie Brilliant Blue R-250) staining (20 applications), 2 μL /lane for silver staining (100 applications).

2) Positive control 3 for western blotting / immunostaining: 2 μL / lane (100 applications).

Optimal working concentrations must be determined by the end user.

NOTES:

1) Stainability of the lower molecular weight bands by immunostaining may be less than that of the higher bands.

2) Stainability of the bands by immunostaining may be different according to animal species and subclass of antibody used and / or staining conditions (see table).

3) Do not use as a blocking reagent.

4) This product is not a prestained marker.

Purified by affinity chromatography on an IgG column. Liquid in SDS-PAGE sample buffer 2 [62.5 mM Tris-HCl (pH 6.8) - 2% SDS - 5% glycerol - 0.005% Bromophenol Blue]. 2 μL of this product contains 0.05 μg each of pA(AB)1-6. Assure that contents are dissolved completely prior to use.

Store at -20°C for up to 1 year from date of purchase. After opening maintain at -20°C in undiluted aliquots for up to 3 months. Avoid repeated freeze/thaw cycles.
