Description
Millipore's ProSep range of media is designed for high throughput affinity purification of monoclonal antibodies and other proteins. Affinity chromatography is elegantly simple and extremely efficient as the first capture step in the purification process of a protein from cell culture.
ProSep® Ultra Plus Chromatography Media
The highest dynamic binding capacity protein A affinity chromatography media, designed for cost effective, large-scale purification of todays higher titer therapeutic antibodies.
ProSep Ultra Plus is a protein A based affinity resin with the highest dynamic binding capacity and flow rate capability of any comparable resin on the market. Based on the proven technology of Prosep, ProSep Ultra Plus media provides increased capacity and productivity compared to competing resin-based technologies. In addition, its rigid base matrix enables predictable scale-up and greater process flexibility, presenting antibody producers with the opportunity to reduce the equipment footprint and purify large volumes of higher titer feedstock in a shorter period of time.

ProSep®-vA Chromatography Media
ProSep-vA chromatography media are designed to facilitate highly efficient, cost effective purification of monoclonal, polyclonal and engineered antibodies. ProSep-vA Ultra and ProSep-vA High Capacity media are manufactured using no mammalian derived products.
ProSep-vA chromatography media consist of protein A immobilized on porous glass that is permeated by interconnecting pores of uniform and precisely controlled size. ProSep-vA chromatography media exhibit chemical and mechanical stability over a range of conditions such as low pH, exposure to detergents, buffers of varying ionic strengths and many organic solvents.
The ProSep matrix is optimized for large-scale production use and offers significant advantages compared with other available matrices:

ProSep-vA Ultra Media
ProSep-vA Ultra media has been designed to maintain the same basic attributes of ProSep-vA High Capacity but gives an even higher binding capacity. It has been specifically optimized to address the needs of todays higher titer, large volume fermentation feedstocks, providing high productivity and low cost of operation.

ProSep-vA High Capacity Media
ProSep-vA High Capacity media is widely used for the purification of monoclonal antibodies, both approved and in various phases of clinical trials. The larger pore size compared with ProSep-vA Ultra media means that it can also be used for the purification of fusion proteins or conjugates, where the effective molecular weight is larger than 150 kDa.

ProSep-PB Media
ProSep-PB media is designed for the affinity chromatography of a variety of glycoproteins, nucleic acids, and other biomolecules containing 1,2 cis-diol functionality. ProSep-PB media can be used in the purification of glycoproteins, nucleotides and viruses.
ProSep-PB media is composed of a synthetic m-aminophenyl ligand immobilized on controlled pore size glass beads. The non-compressible nature of the media and the low back pressure generated, allows efficient media utilization at all scales.

ProSep-G Media
ProSep-G consists of protein G immobilized to the ProSep matrix. ProSep-G is used for the purification of those antibodies, which do not bind efficiently to protein A matrices, such as rat IgG's and human IgG3. ProSep-G can also be used for the purification of a variety of other antibodies. Typically antibodies bind to ProSep-G over a pH range of 4.0 to 8.0, and bound antibodies are eluted at a pH of between 2.5 and 3.0.
In addition to whole antibodies, ProSep-G has been successfully used for the purification of antibody fragments, typically giving capacities of 3mg/L.

ProSep Thiosorb and Thiosorb M
ProSep Thiosorb and Thiosorb M are prepared by the in situ synthesis of a non biological thiophilic ligand. ProSep Thiosorb products selectively bind protein regions common to all immunoglobulins, and can be used for the purification of antibodies or fragments.
Recovery of active antibody is typically greater than 99% due to the mild binding and elution conditions. This can be advantageous in cases where the antibody molecule is sensitive to low pH.

Activated ProSep
Pre-activated ProSep is suitable for the covalent attachment of proteins and other molecules to form an affinity adsorbent. It is available with two surface chemistries, ProSep 5CHO and ProSep 9CHO. Pore diameters of either 500 Å or 1000 Å are available with either chemistry.
