MitoLight Mitochondrial Permeabilization Detection Kits

The MitoLight Mitochondrial Apoptosis Detection Kit is a quick and easy method for detecting changes in the mitochondrial membrane potential in live cells. Mitochondrial depolarization is a well-characterized event in apoptosis that can be detected using MitoLight. This lipophilic cationic dye stains living cell mitochondria in a membrane potential-dependent fashion. In healthy cells, the dye accumulates and aggregates in the mitochondria and fluoresces red (Em = 585–590 nm). During apoptosis, the electrochemical gradient across the mitochondrial membrane breaks down (presumably due to the formation of additional pores in the membrane), and the dye remains in the cytosol in its monomeric form and fluoresces green (Em = 527–530 nm).

Fluorescence Microscopy

Healthy NIH3T3 cells incubated with MitoLight reagent for 30 minutes at 37°C. Note the presence of red mitochondria, indicating an intact membrane potential. During apoptosis the loss of mitochondrial membrane potential leads to a loss of red color as the dye diffuses out of the mitochondria.


The MitoLight Mitochondrial Apoptosis Detection Kit will work with adherent cells and cells grown in suspension. With cells in suspension, the analysis can be performed by fluorescence microscopy or by flow cytometry; with adherent cells, the analysis must be performed using fluorescence microscopy.
