Cell yield and viability data comparing growth of six cell types in T75 flasks vs. Millicell HY cell culture flasks.
Cell were seeded at 20,000 – 40,000 cells/cm
2 with 0.2 mL/cm
2 media and incubated for 48 h at 37 ºC, 6% CO
2, 95% relative humidity. Cells were washed with either PBS or 0.02% EDTA followed by harvest via enzyme dissociation for 10-15 min at 37 ºC. Cell count and viability measurements were performed using a guava
® easyCyte™ flow cytometer with guava ViaCount
® flex reagent. Theoretical cell yield = (cells recovered from one T75 flask) ÷ 75 cm
2 x (total surface area in cm
2).

Performance Comparison
HY flask produces better cell recovery compared to other multilayer cell culture flasks.
To compare cell yield between Millicell HY flasks and other multilayer cell culture flasks, CHO-k1 cells were seeded, recovered, and counted in experiments shown for High Yield. % cell yield = 100 x actual cell yield divided by theoretical cell yield (calculated for High Yield experiments).
