At the capture phase of purification, the primary goals are; to isolate the product of interest as quickly as possible to minimize product degradation, to reduce the process volume to a manageable scale for subsequent steps, and to maximize yield and economy. These goals are best achieved using a highly selective affinity ligand combined with a support that is optimized for maximum throughput and productivity.

To maximize productivity, adsorptive media must possess both high dynamic capacity and high volumetric throughput capability. More rigid matrices clearly offer the best flow rate capability, but for this superiority to be realized, it must be combined with highly favorable mass transport of target solutes to and from available binding sites. The resulting ability to capture target substances, wash unbound contaminants, and elute the bound product quickly, while using minimal media volume, is the basis for productivity and economic optimization.

Our ProSep range of media is designed for cost effective, high throughput affinity purification of monoclonal, polyclonal and engineered antibodies and are especially suited for large-scale purification of today’s higher titer therapeutic antibodies.

ProSep protein A affinity media are manufactured using no mammalian derived products. ProSep protein A chromatography media consist of protein A immobilized onto porous glass that is permeated by interconnecting pores of uniform and precisely controlled size. ProSep chromatography media exhibit chemical and mechanical stability over a range of conditions such as low pH, exposure to detergents, buffers of varying ionic strengths and many organic solvents.

ProSep-vA High Capacity
ProSep-vA High Capacity media is widely used for the purification of monoclonal antibodies, both approved and in various phases of clinical trials. The larger pore size compared with ProSep-vA Ultra media means that it can also be used for the purification of fusion proteins or conjugates, where the effective molecular weight is larger than 150 kDa.
