Tyrosine phosphorylation of and by transmembrane proteins is one of the key steps in so much of today’s research, from early stage drug screening to signal transduction and regulation of enzymatic activity. Yet studying this step can be challenging. For years, the solution has been to settle on receptor fragments commercialized for pair-wise interactions. However, much information is lost in these experiments since signaling proteins need other ‘team members’ to become fully functional. This activity typically arises from the assembly of a complex signaling team formed near the membrane surface. Analysis of a single protein interacting with a single partner loses the rich, two-dimensional information afforded by the membrane surface.
