Active mTOR
Introducting Active mTor by Millipore
Millipore's team of Upstate kinase experts has added active mTOR (Mammalian Target of Rapamycin) to its collection of high-quality active kinases. Recent studies have linked mTOR (a.k.a. FRAP, RAPT or RAFT) to many important human diseases including cancer, diabetes, obesity, cardiovascular diseases and neurological disorders.
| Product | Catalog Number | |
| mTOR (1352-end) | active | 14-770 |
| mTOR substrate | 1 mg | 12-645-K |
| mTOR substrate | 10 mg | 12-645 |
| mTOR (1362-end) | active + FKBP12 | 14-784 |
mTOR's Known Effects Include Regulation of:
- Cell cycle progression
- Cell growth
- Protein synthesis
- Ribosome biogenesis
- Autophagy
Now Available— Full-length mTOR
Exclusively For KinaseProfiler
Full-length biologically relevant mTOR is now available as part of Millipore’s KinaseProfiler service. Our Upstate kinase experts pioneered commercial kinase profiling in 2000. Today, KinaseProfiler offers 264 kinases for selectivity profiling using the gold standard catalytic radiometric assay format as part of Millipore. The addition of full-length mTOR to the KinaseProfiler panel offers you exclusive access to high-quality, confidential screening of your compounds against this critically important kinase.
KinaseProfiler Service:
- Featuring 264 kinases
- Gold-standard catalytic radiometric assays
- High-quality, confidential screening of compounds
Domain Architecture of Full-length And Truncated Forms of mTOR
Functional domains of mTOR: HEAT (Huntington elongation factor 1A-protein phosphatase 2A-A subunit-TOR); FAT (FRAP, ATM, TTRAP2); FRB (FKBP12 rapamycin binding); PIKK (PI3-kinase-related kinase); RD (regulatory domain); and FATC (FAT, C terminal).
The amino acid residue number (top) shows the relative positions of the domains.
IC50 determination for Rapamycin in the presence of 10 µM FKBP12.
Rapamycin was titrated in a semi-logarithmic serial dilution using 100% DMSO
and tested against the two forms of mTOR in the presence of 10 micromolar FKBP12.
Data were converted to % activity and fitted to a four-parameter logistic using GraphPad Prism™. IC50 values for the full-length and truncated forms are shown in Table 1. This complex inhibits full-length and truncated mTOR to similar degrees.
| Table 1 | ||
| Construct | [FKBP12] | IC50 |
| mTOR-(hu, FL) | 10 μm | 25 nM |
| mTOR-(hu, 1362-end] | 10 μm | 20 nM |
