Live Cell Imaging - Mammalian

M04S 4-Color Switching: Switching between 4 fluorescent solutions - Solution exchange in the M04S microfluidic plate between PBS and 4 fluorescent dyes with a system set point of 4 psi. Note the rapid and uniform laminar exchange profile. Movie taken with a 4X objective lens, showing the entry portion of the 2.8 mm diameter culture chamber.	M04S Cell Loading - Cell loading using capillary flow - Loading of MCF-7 cells (2.5 million cells/ml) into the M04S microfluidic culture region via capillary flow. Note that as the cells enter the culture area, they settle onto the glass surface.
M04S Perfusion Culture - MCF-10A cells - Microfluidic culture of MCF10A human breast cancer cells using the CellASIC^™ ONIX microincubation system.	M04S Perfusion Culture - NIH3T3 cells - Microfluidic culture of NIH3T3 mouse fibroblasts using the ONIX microincubation system. Cells exposed to flow of DMEM at 1psi, 37C and 5% CO2, using a 40X objective lens.
M04S Perfusion Culture - HT-1080 cells - HT-1080 cancer cells cultured in the M04S plate with the micro-incubator system imaged with a 40X phase contrast objective for 1 hour.	M04L Solution Switching - Solution switching flow profile - Flow switching in the M04L microfluidic plate taken with a 4X objective. Solution swiching is shown between PBS and Texas Red conjugated dextran (red) at 4 psi. The chamber diameter is 2 mm.
M04G Gradient Switching - Creating and switching a stable concentration gradient - Switching of concentration gradients in the M04G microfluidic plate. Flow of Fluorescein conjugated dextran, 3kDa (green) and PBS at 1psi creates a stable diffusion gradient between the top and bottom channels. Images taken with a 4x objective lens.	M04G Neutrophil Migration - HL-60 cells migrating in response to a spatial gradient - Chemotaxis of differentiated neutrophil-like HL-60 cells in a stable gradient of formyl-Met-Leu-Phe, a potent chemoattractant for granulocytes. M04G HL-60 Migration - HL-60 cells responding to a gradient - HL-60 migration in response to a chemokine (traced with red fluorescence). Unattached cells flow out to the left. M04S Transfection - HT-1080 cells in microfluidic culture tranfected with GFP-tubulin - HT-1080 cells cultured in the M04S microfluidic plate using the microincubation system. Cells were transfected with GFP-tubulin (Invitrogen CellLight) via continuous flow overnight, and imaged at 40X for 1 hour with flow at 1 psi.
M04L Perfusion Culture - Live cell imaging of MCF7 cells - Microfluidic culture of MCF7 human breast cancer cells the M04L with flow at 0.5 psi. 20X objective.	M04S Bacteria Infection - HT-29 cells exposed to an invasive strain of E. coli - The human HT-29 colorectal adenocarcinoma cell line infected with an invasive strain of E. coli. M04S Removal of Cells from Chamber - NIH3T3 Fibroblast cells grown to confluency in the M04S plate. The chamber is treated with trypsin and cells are flushed out by pneumatic pressure thereafter.
