Most small molecule kinase inhibitors are ATP-competitive. The ATP-binding pocket is well-conserved in the protein kinase family making it likely that inhibitors will cross-react with unintended targets.By profiling against the largest panel of protein kinases, a detailed understanding of selectivity can be determined so there are no surprises along the path of drug development. KinaseProfiler Assay
Assay format used in KinaseProfiler. This assay system is a direct enzymatic measurement, which is both robust and reliable — it is the gold standard radiometric filter binding format.

Initial screening is performed at either a single, customer specified concentration or an IC50 analysis can be performed on selected compounds.
* Homogenous time resolved fluorescence (HTRF) technology is utilized for PI 3-kinases that have been configured into a robust homogenous format.
Assay Flexibility and Compatibility
To ensure that data generated by Millipore is comparable with existing and future client-generated data, we offer flexibility in the assay parameters whereby many can be selected and/or modified based on client requirements. These include:
- replicate numbers
- number of kinases tested
- number of compound concentrations
- ATP concentrations used in the assay
Convenience, Speed and Confidentiality
Compounds can be supplied by clients as DMSO stocks of known concentration in either vials or 96-well plates, or as solid materials in vials. If the KinaseProfiler service is needed on a continued basis, assay slots can be reserved allowing a steady flow of lead compounds to be subjected to the fastest possible throughput.
